摘要
为了探究不同温度条件下添加卵黄对猪精子微胶囊质量参数及其缓释率的影响,试验在配制Modena液时添加20%卵黄,同时制备精子微胶囊。使用全自动精子分析仪检测17℃、37℃温度条件下72 h内胶囊组、卵黄组和正常组精液质量,以及在37℃温度条件下48 h内无卵黄组和有卵黄组精子微胶囊缓释率。结果表明:在17℃温度条件下,卵黄组精子活率在6~24小时时显著高于胶囊组和正常组(P<0.05);精子畸形率在1~6小时时显著低于胶囊组(P<0.05),在3~6小时时与正常组比较差异不显著(P>0.05);精子路径速度在3~72小时时显著高于胶囊组(P<0.05),在36~72小时时显著高于正常组(P<0.05);精子直线速度在36~72小时时显著高于胶囊组(P<0.05),在36~48小时时与正常组比较差异不显著(P>0.05);精子向前性在72小时时显著高于正常组(P<0.05)。在37℃温度条件下,各组精子质量参数均差异不显著(P>0.05);有卵黄组的缓释率均高于无卵黄组,其中在4,6,48小时时差异显著(P<0.05)。说明添加20%卵黄能够提高精子微胶囊质量参数及其缓释率。
In order to explore the effect of adding yolk at different temperatures on the quality parameters and sustained release rate of porcine sperm microcapsules, 20% egg yolk was added in the basis of Modena basic diluent, and sperm microcapsules were prepared at the same time. The semen quality in capsule group, egg yolk group and normal group within 72 h at 17 ℃ and 37 ℃ and the microcapsule sustained release rate of sperm in non-egg yolk group and egg yolk group within 48 h at 37 ℃were detected by Mailang sperm analysis system. The results showed that the sperm viability in egg yolk group was significantly higher than that in capsule group and normal group at 17 ℃ for 6-24 h(P<0.05). The sperm malformation rate in egg yolk group was significantly lower than that in capsule group at 1-6 h(P<0.05), and there was no significant difference between egg yolk group and normal group at 3-6 h(P>0.05). The sperm path velocity in egg yolk group was significantly higher than that in capsule group at 3-72 h(P<0.05), and was significantly higher than that in normal group at 36-72 h(P<0.05). The linear velocity of sperm in egg yolk group was significantly higher than that in capsule group at 36-72 hours(P<0.05), but there was no significant difference compared with normal group at 36-48 h(P>0.05). Sperm progressivity at 72 h was significantly higher than that in the normal group(P<0.05).At 37 ℃, there was no significant difference in data profiles among all groups(P>0.05);the sustained release rate in egg yolk group was higher than that in the non-egg yolk group, and the difference was significant at 4, 6 and 48 hours(P<0.05). The results indicated that adding 20% egg yolk could improve the quality parameters of sperm and increase the sustained release rate of microcapsules.
