细粒棘球绦虫Calmodulin蛋白的生物信息学分析 被引量:2

摘要 目的预测细粒棘球绦虫钙调蛋白(Echinococcus granulosus calmodulin, EgCalmodulin)的生化特性、结构和抗原表位,为包虫病分子肽疫苗的筛选奠定基础。方法利用在线数据库以及生物信息学相关软件分析EgCalmodulin蛋白的理化性质、二级和三级结构、亲/疏水性、表面可及性、抗原指数和柔性区域以及该蛋白的抗原表位。结果细粒棘球绦虫Calmodulin蛋白是由118个氨基酸组成,相对分子质量为13.48×10^(3),属于较稳定蛋白质;不含信号肽序列和跨膜结构域;二级结构中α-螺旋占47.46%,β-转角占13.56%,延伸连占15.25%,无规则卷曲占23.73%;亲水区域明显,评分较高的抗原指数所在区域与柔性区域相对应;优势B细胞表位位于10-20、32-37、47-55、65-74、81-91、100-110氨基酸区段;优势T细胞表位位于99-105、33-41、87-95,45-56、99-110、9-21氨基酸区段。结论利用生物信息学方法预测EgCalmodulin蛋白含有优势B、T细胞表位,可为包虫病分子肽疫苗的筛选提供参考。 Objective To predict the biochemical characteristics, structure, and epitopes of Echinococcus granulosus calmodulin in order to lay the foundation for the screening of molecular peptide vaccines against echinococcosis. Methods The physicochemical properties, secondary and tertiary structures, hydrophilicity/hydrophobicity, surface accessibility, antigenic index, flexible regions, and epitopes of the E. granulosus protein calmodulin were analyzed using online databases and bioinformatic software. Results The calmodulin protein of E. granulosus consists of 118 amino acids with a relative molecular weight of 13.48 ×10^(3). Calmodulin is a relatively stable protein. It contains no signal peptide sequences or transmembrane domains. Its secondary structure consists of α-helices(47.46%), β-turns(13.56%), extended strands(15.25%), and random coils(23.73%). The hydrophilic region was obvious, and the region with a higher antigenic index corresponded to the flexible region. Dominant B-cell epitopes were located in amino acid domains 10-20, 32-37, 47-55, 65-74, 81-91, and 100-110. Dominant T-cell epitopes were located in amino acid domains 99-105, 33-41, 87-95, 45-56, 99-110, and 9-21. Conclusion Bioinformatics was used to predict that the E. granulosus protein calmodulin contains dominant B-and T-cell epitopes. This finding can serve as a reference for the screening of molecular peptide vaccines against hydatid disease.
