摘要
目的通过对丹参-红花配方颗粒与传统汤剂的主要成分及药效进行研究,评价配方颗粒与传统汤剂在化学成分和药理效应方面的一致性。方法采用多指标定性、定量相结合的方法对配方颗粒和传统汤剂的主要化学成分进行分析;采用急性心肌缺血模型大鼠对丹参-红花配方颗粒和传统汤剂的药效进行初步评价。结果配方颗粒和汤剂TLC图的主斑点清晰、圆整、分离度良好,且数目一致;配方颗粒剂中除迷迭香酸外,丹参素、丹酚酸B和紫草酸的质量浓度均高于汤剂,方法学考察符合含量测定的要求;丹参-红花配方颗粒及汤剂的心肌病理切片显示,心肌纤维排列整齐,细胞核清晰,炎症细胞浸润减少,对缺血心肌具有良好改善作用。结论丹参-红花配方颗粒及汤剂的主要成分的种类和对缺血心肌的改善作用基本一致,颗粒剂中主要成分的质量浓度均高于汤剂。
Objective To evaluate the consistency of chemical composition and pharmacological effects of dispensing granules and traditional decoction by studying the main components and pharmacological effects of Danshen-Honghua dispensing granules and traditional decoction.Methods The main chemical components of dispensing granules and traditional decoction were analyzed by a combination of multiple indicators,qualitative and quantitative methods.The efficacy of Danshen-Honghua dispensing granules and traditional decoction was evaluated by acute myocardial ischemia model rats.Results The main spots of TCL map of the dispensing granules and the decoction were clear,round and well separated,and the number of main spots was the same.Except for rosmarinic acid,the contents of Danshensu,salvianolic acid B and shikonic acid in the granules were higher than those of the decoction,and its methodology met the requirements for determination.The myocardial pathological section of Danshen-Honghua dispensing granules and decoction had neatly arranged myocardial fibers,clear cell nuclei,and reduced inflammatory cell infiltration,which had a good effect on the ischemic myocardium.Conclusion The types of the main components of the Danshen-Honghua dispensing granules and the decoction are basically the same as the improvement effect on ischemic myocardium,and the content of the main components in the granules is higher than that of the decoction.
