核桃分心木水提物抗疲劳作用研究 被引量:6

摘要 目的:研究核桃分心木水提物的抗疲劳作用,为其保健品开发和应用提供科学依据。方法:制备分心木水提物并测定其总多糖、总多酚和总黄酮含量。将小鼠随机分为空白对照组和分心木水提物低、中、高剂量组,连续灌胃14 d,测定小鼠的体重、脏器指数和负重游泳力竭时间,检测血清乳酸(BLA)、乳酸脱氢酶(LDH)、尿素氮(BUN)、肌肉和肝脏糖原含量,以及肝脏丙二醛(MDA)含量和超氧化物歧化酶(SOD)活性。结果:分心木水提物得率为(9.41%±0.82%),总多糖、总多酚、总黄酮含量分别为(33.40%±0.88%)、(30.80%±2.28%)、(24.62%±0.86%)。分心木水提物对小鼠的体重和脏器指数没有明显影响(P>0.05),能延长小鼠的负重游泳力竭时间(P<0.01),并具有剂量依赖性,能降低小鼠BLA和BUN含量,提高LDH活性(P<0.01)。与空白对照组相比,给药组小鼠运动后肝、肌糖原含量增加,肝脏MDA含量降低、SOD活性升高(P<0.01)。结论:分心木水提物能提高小鼠的运动耐力,具有抗疲劳作用。 Objective To determine the anti-fatigue activity of aqueous extract from Diaphragma juglandis Fructus(AED)in walnut,and set base for further application of the extract.Method Diaphragma juglandis Fructus was extracted with water under reflux.The extracting liquid was concentrated and lyophilized.The total polysaccharides,polyphenol and flavonoid in AED were measured.Mice were randomly divided into control group,low dose AED group,middle dose AED group and high dose AED group.After AED was orally administered to mice continuously for 14 days,the body weight,organ index,load swimming time,blood lactic acid(BLA),blood urea nitrogen(BUN),serum lactate dehydrogenase(LDH),hepatic and muscle glycogen,hepatic malondialdehyde(MDA)and superoxide dismutase(SOD)activity of the mice were measured to evaluated the anti-fatigue effect of AED.Result The yield rate of the lyophilized AED was(9.41%±0.82%).The total polysaccharides,polyphenol and flavonoid in AED were(33.40%±0.88%),(30.80%±2.28%)and(24.62%±0.86%)differently.There was no difference among the groups about body weight and organ index(P>0.05).AED could prolong the exhaustive swimming time,lower the content of BLA and BUN,increase serum LDH activity(P<0.01).Compare to the control group,hepatic and muscle glycogen,hepatic SOD activity of the mice in the three dose groups after exercise increased,while hepatic MDA content decreased(P<0.01).Conclusion AED could increase endurance capacity of physical exhaustion and had anti-fatigue activity.
