摘要
试验对新疆南部5个地区密点麻蜥线粒体DNA(Mitochondrial DNA,mtDNA)细胞色素b(Cytochrome b,Cytb)基因序列进行PCR扩增和测序,采用邻接法和最大似然法分别构建系统进化树进行系统发育分析。结果表明:南疆沙蜥独立于1个分支;所有麻蜥在1个分支上,与山地麻蜥(Eremias brenchleyi)的亲缘关系最远,与荒漠麻蜥(Eremias przewalskii)的亲缘关系最近;密点麻蜥种群间每个地区样本几乎均是独立分支,喀什地区的密点麻蜥与民丰的密点麻蜥亲缘关系最远,乌恰次之,与同市叶城和塔什库尔干的密点麻蜥亲缘关系最近,但喀什地区的M13稍有偏离,与乌恰的密点麻蜥属于同1个分支,表明这两地区的密点麻蜥存在基因交流。文章基于Cytb基因对新疆南部5个地区密点麻蜥进行系统发育分析,补充和丰富了新疆部分地区密点麻蜥的基础遗传数据。
The research carried out PCR amplification and sequencing on the mitochondrial DNA(mtDNA)cytochrome b(Cytb)gene sequence of the Eremias multiocellata in five regions of southern Xinjiang.The phylogenetic tree was constructed by neighbor-joining(NJ)and maximum likelihood(ML)methods for phylogenetic analysis.The results showed that the lizard was independent of one branch.All lizards belong to the same branch,and the most closely related to the mountain lizards and the most closely related to the desert lizards.Eremias multiocellata each region between samples are almost independent branch of kashgar region of Eremias multiocellata and freeman of kinship,as far as the point of hemp lizard just take second place,with City Ye and Tashikuergan thick hemp lizard's closest relative,but the M13 slightly deviate from kashgar region,and just the size of hemp lizards belonging to the same branch,which suggests that there was gene exchange between the two species.Based on Cytb gene,phylogenetic analysis of the lizard in five areas of southern Xinjiang was conducted in the study,which supplemented and enriched the basic genetic data of the lizard in some areas of Xinjiang,and provided a certain theoretical basis for conservation and rational utilization of germplasm resources in the later stage.
