摘要
本实验以高糖自交系西州蜜(X25)和低糖自交系伽师瓜(JS)为亲本构建F2群体XJ F2,以高糖品种网纹甜瓜M157自交系和低糖自交系伽师瓜(JS)为亲本构建F2群体MJ F2。通过分析两个群体的果实糖度频率分布,发现两个群体的果实糖度均呈正态分布,符合数量性状遗传的典型特征。利用SSR标记通过集群分离分析法对糖含量性状相关的基因进行初步定位,发现XJ F2群体中具有3个SSR标记ECM134(LG4),MU3349(LG11)和CMAGN45(LG11)与甜瓜果实的糖含量性状连锁,并在MJ F2群体中发现两个SSR标记MU5035(LG10)和chr10-0380(LG10)与甜瓜果实的糖含量性状连锁。通过对已知甜瓜糖代谢相关酶基因在基因组中位点分析,发现与果实糖含量性状相关的位点与基因组中糖代谢酶基因存在共定位关系。本研究结果为后续对与果实糖含量性状相关基因的定位奠定了基础。
In this experiment,the high-sugar inbred lines Xizhou honey(X25)and the low sugar inbred line Jiashi melon(JS)were used as parents to construct F2 population XJ F2.High-sugar varieties reticulated melon inbred line M157 and low-sugar inbred line JS were used as parents to construct F2 population MJ F2.By analyzing the freq-uency distribution of fruit sugar degree of the two populations,it was found that the fruit sugar degree of the two populations were normal distribution,which accorded with the typical characteristics of quantitative character inh-eritance.SSR markers were used to map genes related to sugar content traits by bulked segregation analysis(BSA).The results showed that three SSR markers ECM134(LG4),MU3349(LG11)and CMAGN45(LG11)were found to be linked to sugar content traits in melon fruits in XJ F2 and two SSR markers MU5035(LG10),chr10-0380(LG10)were found to be linked to the sugar content traits of the melon fruit in MJ F2.Through the locus analysis of the known genes of sugar metabolism related enzymes in melon genome.It was found that the loci related to the sugar content of fruit were co-localized with the gene of sugar metabolism enzyme in genome.This study laid a foundation for the location of genes related to sugar content in fruit.
