Study on Nrf2/HO-1 Pathway and Oxidative Stress-mediated Pulmonary Toxicity of Ethanol Extract from Aboveground Parts of Chloranthus Serratus

摘要 目的:探索及己地上部分醇提取物对大鼠的肺毒性及其机制.方法:取24只雄性SD大鼠随机分为空白组(KB)、DC低组(50倍)以及DC高组(100倍).提取物组灌胃给予不同剂量及己地上部分醇提取物混悬液,KB组用同等剂量的0.5%羧甲基纤维素钠溶液灌胃,给药14天,计算大鼠体重变化率和肺脏器系数;检测肺匀浆中氧化应激指标含量;苏木精-伊红染色法考察肺组织病理变化;免疫组化法和Western blotting法分别检测ICAM-1阳性表达水平和Nrf2/HO-1蛋白表达情况.结果:与KB组相比,DC高组体重增长率较低.提取物组肺表面有明显出血点,且DC高组更明显.光镜下DC高组支气管炎性浸润较多,管壁纤维紊乱呈锯齿状增生,血管壁有纤维淀粉样变性和出血,DC高组症状较DC低组明显.与KB组相比,DC高组T-SOD、GSH和CAT水平显著性降低,MDA水平极显著性升高,DC低组氧化应激指标无统计学差异;提取物组ICAM-1阳性表达显著性增加,而Nrf2和HO-1蛋白表达量显著性下降,且DC高组较DC低组更明显.结论:及己地上部分醇提取物高剂量组致大鼠肺毒性大于低剂量组,其机制可能与抑制Nrf2/HO-1通路及氧化应激有关. Objective:To explore the pulmonary toxicity of ethanol extract of aboveground parts of Chloranthus serratus in rats and its mechanisms.Methods:24 male SD rats were randomly divided into blank group(KB),low DC group(50 times)and high DC group(100 times).The KB group was given the same dose of 0.5%sodium carboxymethylcellulose solution intragastrically for 14 days.Results:Compared with the KB group,the weight growth rate of the DC high group was lower.There were obvious bleeding spots on the lung surface in the extract group,and more obvious in the DC high group.Under the light microscope,the DC high group had more bronchial inflammatory infiltration,the disordered fiber in the wall of the tube shows a serrated hyperplasia,and the blood vessel wall had fibrous amyloidosis and hemorrhage.The symptoms of the DC high group were more obvious than those of the low DC group.Compared with the KB group,the levels of T-SOD,GSH and CAT in the DC high group were significantly reduced,and the level of MDA increased significantly.There were no statistical differences in the oxidative stress indicators in the DC low group,the positive expression of ICAM-1 in the extract group significantly increased,but Nrf2 and HO-1 protein expression decreased significantly,and the DC high group was more obvious than the DC low group.Conclusion:The high-dose group of ethanol extract of aboveground part of C.serratus caused greater pulmonary toxicity in the rats than the low-dose group.The mechanisms may be related to the inhibition of the Nrf2/HO-1 pathway and oxidative stress.
