虎杖苷对脂多糖介导的皮肤成纤维细胞焦亡的影响

摘要 目的探讨成纤维细胞焦亡在创面愈合中的作用及虎杖苷对其的影响。方法以5μg/ml脂多糖(LPS)刺激成纤维细胞建立实验模型,并随机分为4组:对照组细胞不接受任何处理;实验组细胞接受5μg/mlLPS处理;治疗组细胞接受5μg/mlLPS及50μmol/L虎杖苷处理;抑制剂组细胞接受5μg/mlLPS及100 nmol/LVX-765处理。四甲基偶氮唑蓝(MTT)法检测细胞增殖;试剂盒检测caspase-1活性;酶联免疫吸附试验(ELISA)检测培养基白细胞介素(IL)-1β、IL-18及Ⅲ型胶原蛋白含量。结果与对照组比较,实验组caspase-1活性及IL-1β、IL-18含量分别显著增加为(475.4±39.9)%、(254.9±19.4)pg/ml及(212.6±22.4)pg/ml,细胞增殖及Ⅲ型胶原蛋白含量分别显著下降为(72.5±3.4)%及(0.06±0.01)μg/L(均P<0.05);与实验组比较,治疗组caspase-1活性及IL-1β、IL-18含量分别显著下降为(292.1±28.5)%、(167.5±11.8)pg/ml及(165.5±19.9)pg/ml,细胞增殖及Ⅲ型胶原蛋白含量分别显著增加为(86.8±5.5)%及(0.10±0.02)μg/L(均P<0.05);与实验组比较,抑制剂组caspase-1活性及IL-1β、IL-18含量分别显著下降为(185.5±16.1)%、(127.4±16.5)pg/ml及(129.5±17.2)pg/ml,细胞增殖及Ⅲ型胶原蛋白含量分别显著增加为(83.1±7.7)%及(0.09±0.01)μg/L(均P<0.05)。结论抑制LPS诱导的成纤维细胞焦亡可以促进其增殖并分泌胶原蛋白,虎杖苷可能通过抑制成纤维细胞焦亡促进创面愈合。 Objective To investigate the effect of polydatin on the pyroptosis of fibroblasts in wound healing.Methods Fibroblasts were stimulated with 5μg/ml of lipopolysaccharide(LPS)to establish the experimental model,and were randomly divided into four groups as following:cells in the control group received none treatment;cells in the experimental group were treated with 5μg/ml of LPS;cells in the treatment group were treated with 5μg/ml of LPS and 50μmol/L of polydatin;cells in the inhibitor group were treated with 5μg/ml of LPS and 100 nmol/L of Belnacasan(VX-765).Cell proliferation was detected by 3-(4,5)-dimethylthiahiazo(-2)-3,5-diphenytetrazoliumromide(MTT)assay.Caspase-1 activity was detected by the commercial kit.The contents of interleukin(IL)-1β,IL-18,and type Ⅲ collagen in the culture medium were determined by enzyme linked immunosorbent assay(ELISA).Results Compared with those in the control group,the caspase-1 activity and IL-1β and IL-18 contents in the experimental group significantly increased to(475.4±39.9)%,(254.9±19.4)pg/ml,and(212.6±22.4)pg/ml,respectively,and the cell proliferation and type Ⅲ collagen content significantly decreased to(72.5±3.4)%and(0.06±0.01)μg/L(all P<0.05).Compared with those in the experimental group,the caspase-1 activity and IL-1β and IL-18 contents in the treatment group decreased significantly to(292.1±28.5)%,(167.5±11.8)pg/ml,and(165.5±19.9)pg/ml,respectively,and the cell proliferation and type Ⅲ collagen content significantly increased to(86.8±5.5)%and(0.10±0.02)μg/L(all P<0.05).Compared with those in the experimental group,the caspase-1 activity and IL-1β and IL-18 contents in the inhibitor group significantly decreased to(185.5±16.1)%,(127.4±16.5)pg/ml,and(129.5±17.2)pg/ml,respectively,and the cell proliferation and type Ⅲ collagen content significantly increased to(83.1±7.7)%and(0.09±0.01)μg/L(all P<0.05).Conclusion Inhibition of LPS-induced pyroptosis promotes the proliferation and secretion of collagen of fibroblasts.Polydatin may promote the wound healing by inhibiting the pyroptosis of fibroblasts.
