外源性激素对光棘球海胆性别相关基因dmrt1、foxl2和boule表达的影响

Effects of Exogeneous Hormone on Expression of dmrt1,foxl2 and boule Genes in Gonads of Sea Urchin Mseocentrotus nudus

摘要 将质量为(25.04±1.75)g的光棘球海胆Mseocentrotus nudus随机分为三组,放入室内用网隔成3个区域的1000 L水槽:每天用微量注射器由围口膜处注射10μL雌二醇应用液(雌二醇组)、睾酮应用液(睾酮组)和葵花籽油(对照组)。持续注射14 d后,收集海胆的体腔液,分离体腔液上清,检测激素含量;收集海胆的消化道和部分性腺组织用酶联免疫吸附技术、组织切片和酶标仪等检测其激素含量、性腺发育及性腺中dmrt1、foxl2和boule的表达水平变化。结果表明:光棘球海胆精巢中睾酮的含量是卵巢中的2.99倍;卵巢中雌二醇和孕酮的含量显著高于精巢。经雌二醇处理,海胆精巢中dmrt1基因的表达量较对照组显著降低70.8%。经睾酮处理,卵巢中foxl2和boule基因的表达量显著上调,而精巢中dmrt1和boule基因的表达量分别降低72.6%和71.0%。推测,雌二醇和睾酮可能在光棘球海胆性腺发育中发挥重要的调控作用。 Sea urchin(Mseocentrotus nudus)with body weight of(25.04±1.75)g were put into a 1,000 L water tank separated into three parts by a net and randomly divided into three groups:the sea urchin injected with estradiol(E2)application solution(estradiol group),testosterone(T)application solution(testosterone group)and sunflower oil(control group)at a dose of 10μL at perioral membrane by a micro syringe every day.After continuous injection for 14 days,the body fluid of the sea urchin was collected,and the supernatant of the body cavity fluid was separated for detection of the hormone contents;the digestive tract and part of the gonad of the sea urchin were sampled to detect the hormone contents,gonadal development and expression levels of dmrt1,foxl2 and boule in gonads by enzyme-linked immunosorbent assay,real-time quantitative PCR,histological technique and a microplate reader.The results showed that there were gender differences in the content of E2,T and progesterone(PROG)in the gonads of the sea urchin,as 2.99-fold in testis as that in ovary,in which the concentrations of E2 and PROG were 1.87 and 2.35 times as that in the testis,respectively.There was significantly decrease by 70.8%in the expression of dmrt1 gene in testis compared with the control group after continuous injection of E2.In ovary,however,the expression level of foxl2 and boule genes were significant increased after E2 treatment.Furthermore,the expressions of the genes were significantly decreased in testis after MT treatment by 72.6%in dmrt1 gene and by 71.0%in boule gene.In conclusion,E2 and MT are involved in the regulation of gonad development in the sea urchin.
