摘要
目的观察强肝胶囊对猪血清诱导大鼠肝纤维化的治疗作用。方法用猪血清建立大鼠肝纤维化模型后,给予强肝胶囊治疗肝纤维化大鼠。使用HE染色检测肝组织病理学变化;称重并计算肝脏、脾脏、胸腺指数,使用试剂盒检测肝功能情况:丙氨酸氨基转移酶(alanine aminotransferase,ALT)、天冬氨酸氨基转移酶(aspartate aminotransferase,AST)、γ-谷氨酰转移酶(gamma-glutamyl transpeptidase,γ-GT)、丙二醛(malondialdehyde,MDA),酶联免疫吸附试验(enzyme linked immunosorbent assay,ELISA)检测肝纤维化情况:层黏连蛋白(laminin,LN)、透明质酸(hyaluronic acid,HA)、Ⅰ型胶原蛋白(collagen typeⅠ,Col-Ⅰ)、前胶原氨基Ⅲ型端肽(procollagenⅢN-termi,PCⅢ)、Ⅳ型胶原蛋白(typeⅣcollagen,Ⅳ-C)及转化生长因子-β1(transforming growth factor-β1,TGF-β1)。结果强肝胶囊使肝组织纤维间隔变细变短,减少了肝纤维化组织中ALT、AST、γ-GT的含量(P<0.05),减少了肝纤维化大鼠血清及肝脏组织中LN、HA、Col-Ⅰ、PCⅢ、Ⅳ-C、TGF-β1的含量(P<0.05)。结论强肝胶囊对大鼠肝纤维化有一定的治疗作用。
Objective To investigate the therapeutic effects of Qianggan capsule on hepatic fibrosis in rat induced by porcine serum.Methods The rat models with liver fibrosis were established with porcine serum,then the rats were treated by Qianggan capsule.HE stain was used to detect the pathological changes of liver tissue,weighing and calculating the index of liver,spleen and thymus,and the liver function indexes were detected,including alanine aminotransferase(ALT),aspartate aminotransferase(AST),gamma-glutamyl transpeptidase(γ-GT),malondialdehyde(MDA).Moreover,the liver fibrosis indxes including laminin(LN),hyaluronic acid(HA),collagen typeⅠ(Col-Ⅰ),procollagen amino type III telopeptide(PCⅢ),collagen typeⅣ(Ⅳ-C)and proliferating factor-beta 1(TGF-β1)were detected by enzyme linked immunosorbent assay(ELISA).Results Qianggan capsule can shorten the fibrous septum of liver tissue,decrease the content of ALT,AST,andγ-GT in liver tissues of rats,and redice the levels of LN,HA,Col-Ⅰ,PCⅢ,Ⅳ-C,TGF-β1 in serum and liver tissues of the rats.Conclusion Qianggan capsule has certain therapeutic effects on hepatic fibrosis in rats.
