摘要
目的:观察标本配穴电针对腹泻型肠易激综合征(IBS-D)模型大鼠一般体征、血清中TNF-α、INF-γ、SP含量水平、内脏高敏感性及肠组织形态学的改变,探讨标本配穴电针对腹泻型肠易激综合征大鼠的作用机制。方法:本实验采用SPF级雄性Wistar大鼠40只,体质量180~220 g,编号后随机分空白组、模型组、标本配穴组和西药组,每组各10只。空白组正常饲养,其余3组以急性应激与慢性应激相结合的方法制备IBS-D大鼠模型。标本配穴组以电针足三里、关元和内关穴方法治疗;西药组每日予以15 mg/kg匹维溴铵混悬液灌胃进行治疗。结果:与空白组相比,模型组、标本配穴组及西药组的粪便含水量升高,差异具有统计学意义(P<0.05),治疗后标本配穴组和西药组较模型组粪便含水量降低,差异具有统计学意义(P<0.05);除空白组外,其他3组内脏敏感性增高,差异具有统计学意义(P<0.05),治疗后标本配穴组和西药组较模型组内脏高敏感性降低,差异具有统计学意义(P<0.05);与空白组相比,其他3组血清中TNF-α、INF-γ、SP和D-乳酸含量增高,差异具有统计学意义(P<0.05),治疗后标本配穴组和西药组较模型组血清中TNF-α、INF-γ、SP和D-乳酸含量降低,差异具有统计学意义(P<0.05);与空白组相比,其他3组大鼠肠粘膜超微结构损伤,治疗后标本配穴组和西药组较模型组大鼠肠粘膜超微结构完整。结论:标本配穴电针可以改善IBS-D大鼠一般体征、内脏高敏感性及肠组织形态学。
Objective:To observe the changes in general signs,serum levels of TNF-α,INF-γ,SP and D-lactate,visceral hypersensitivity and intestinal histomatology of rats with diarrhea predominant irritable bowel syndrome(IBS-D),and to explore the mechanism of primary-secondary acupoints association electro-acupuncture in treating IBS-D.Methods:40 SPF Wistar rats were randomly divided into the blank group,the model group,and electro-acupuncture group and the western medication group,with 10 rats in each group.IBS-D model was established by the combination of acute stress and chronic stress.The electro-acupuncture group was electro-needling Zusanli(ST36),Guanyuan(CV4)and Neiguan(PC6)with the primary-secondary acupoints association,and the western medication group was treated with Pivemonium Bromide Suspension(15 mg/kg).Results:The fecal water content was significantly increased in the model group,the electro-acupuncture group and the western medication group compared to that in the blank group after modeling(P<0.05);the fecal water content was greatly decreased in the electro-acupuncture group and the western medication group compared to that in the model group after the treatment(P<0.05).The visceral sensitivity was increased in the model group,the electro-acupuncture and the western medication group compared to that in the blank group after modeling(P<0.05);the visceral sensitivity was lower in the electro-acupuncture group and the western medication group than that in the model group after the treatment(P<0.05).The serum levels of TNF-α,INF-γ,SP and D-lactate were significantly increased after modeling(P<0.05);which were significantly decreased after the interventions of electro-acupuncture and western medication(P<0.05).The ultrastructure of the intestinal mucosa was damaged after modeling,which became more complete after the interventions of electro-acupuncture and western medication.Conclusion:Primary-secondary acupoints association electro-acupuncture can improve general signs,visceral hypersensitivity and intestinal histomorphology of the rats with IBS-D.
