补脑Ⅰ号联合骨髓间充质干细胞对缺氧缺糖脑微血管内皮细胞损伤VEGF、ICAM-1表达的影响 被引量:2

Effects of Bunao No.1 combined with BMSCs on the expression of VEGF and ICAM-1 in oxygen-glucose deprivation brain microvascular endothelial cells injury

摘要 目的:通过观察补脑Ⅰ号、骨髓间充质干细胞(BMSCs)对缺氧缺糖(OGD)损伤小鼠脑微血管内皮细胞(BMVECs)的保护作用,探讨补脑Ⅰ号治疗阿尔茨海默病(AD)可能机制。方法:采用OGD方法构建小鼠BMVECs损伤模型,用补脑Ⅰ号含药血清、BMSCs进行干预,按实验分组给予含相应血清的培养液处理。采用RT-PCR法及Western blot法检测各组补脑Ⅰ号含药血清、BMSCs对各组细胞血管内皮生长因子(VEGF)、细胞间黏附分子-1(ICAM-1)基因及蛋白表达水平。结果:OGD能导致BMVECs中VEGF、ICMA-1基因及蛋白表达增多(P<0.05)。10%含药血清、BMSCs、BMSCs+10%正常血清组、BMSCs+10%含药血清组均可升高VEGF基因及蛋白的表达、降低ICAM-1基因及蛋白的表达(P<0.05);其中,与10%含药血清组、BMSCs组、BMSCs+10%正常血清组比较,BMSCs+10%含药血清组能增加VEGF基因及蛋白表达、降低ICAM-1基因及蛋白表达(P<0.05)。结论:补脑Ⅰ号含药血清、BMSCs可能通过抑制炎症反应、促进VEGF分泌,从而减少BMVEGs损伤达到保护血管作用,且补脑Ⅰ号联合BMSCs应用效果优于单独使用。 Objective: To observe the protective effects of Bunao No.1 and bone marrow mesenchymal stem cells(BMSCs) on brain microvascular endothelial cells(BMVECs) in mice with oxygen glucose deprivation(OGD) injury, and to explore the possible mechanism of Bunao No.1 in the treatment of Alzheimer’s disease(AD). Methods: The model of mice BMVECs injury was established by OGD method. The serum containing Bunao No.1 and BMSCs were intervened, and the culture solution containing corresponding serum was given according to the experimental groups. RT-PCR and Western blot were used to detect the gene and protein expression of vascular endothelial growth factor(VEGF) and intercellular adhesion molecule-1(ICAM-1) in each group. Results: The expression of VEGF and ICMA-1 gene and protein in BMVECs was increased because of oxygen-glucose deprivation(P<0.05). Serum containing Bunao No.1, BMSCs, BMSCs+normal serum group, and serum containing Bunao No.1+BMSCs could increase the expression of VEGF gene and protein, and decrease the expression of ICAM-1 gene and protein(P<0.05);Bunao No.1 medicated serum+BMSCs group could increase VEGF gene and protein expression and decrease ICAM-1 gene and protein expression compared with Bunao No.1 medicated serum group BMSCs+normal serum group,and BMSCs group(P<0.05). Conclusion: Bunao No.1 containing serum and BMSCs may protect blood vessels by inhibiting inflammatory reaction and promoting VEGF secretion to reduce BMVEGs damage, and the application effect of Bunao No.1 combined with BMSCs is better than that of single use.
