摘要
目的观察复方青风藤巴布剂贴敷给药对佐剂性关节炎大鼠药效学的影响。方法通过左后跖皮内注射完全弗氏佐剂(Complete Freund’s Adjuvant,CFA)建立佐剂性关节炎(Adjuvant Arthritis,AA)大鼠模型,造模后第18天根据足跖肿胀度将40只AA大鼠随机分为模型组、复方青风藤巴布剂高、中、低剂量组12.0、6.0、3.0 g/(kg·d)、阳性药组,每组8只,另取8只SD大鼠作为空白对照组。于造模后第19天在腹部进行贴敷给药,每日1次,连续给药7 d,模型组和空白对照组均贴敷空白基质巴布剂。每日观察大鼠的一般情况、体重和继发侧足爪肿胀度变化,末次给药12 h后运用ELISA法检测血清白细胞介素-1β(IL-1β)和肿瘤坏死因子(TNF-α),HE染色观察关节软骨和滑膜的形态学变化。结果与空白对照组相比,模型组大鼠体重降低,继发侧足肿胀度明显;与模型组相比,复方青风藤巴布剂各剂量组可显著改善CFA诱导的大鼠体重变化,并降低CFA诱发的大鼠继发侧足肿胀度。此外,模型组大鼠血清中IL-1β、TNF-α水平显著升高,大鼠滑膜组织有大量炎性细胞浸润,血管翳形成,关节软骨受损严重;复方青风藤巴布剂各剂量组均可显著降低血清中升高的IL-1β和TNF-α(P<0.05),高剂量组在减轻AA大鼠的滑膜病变和关节软骨损伤方面的作用最为显著。结论复方青风藤巴布剂可对佐剂性关节炎的炎症损伤起到明显治疗作用,可能与其能抑制相关炎性因子水平有关。
Objective To observe the pharmacodynamics of Compound Qingfengteng Cataplasm on adjuvant arthritis rats.Methods Intradermal injection induced adjuvant arthritis(Adjuvant Arthritis, AA) in rats into the left hind metatarsal of Complete Freund’s Adjuvant(Complete Freund’s Adjuvant, CFA).After 18 days of modeling, 40 rats with AA were randomly divided into model group, the high, medium and low dose groups of Compound Qingfengteng Cataplasm 12.0,6.0,3.0 g/(kg·d) and sinomenine gel positive control group according to the degree of plantar swelling.Another eight normal rats were used as blank control group.After 19 days of modeling, the drug was applied to the abdomen once a day for 7 consecutive days, and the model group and the normal group were applied blank matrix cataplasm.After the last administration, the serum interleukin-1β(IL-1β) and tumor necrosis factor-α(TNF-α) were detected by ELISA,and the morphological changes of articular cartilage and synovium were observed by HE.Results Compared with the blank control group, the weight of rats in the model group were decreased and the secondary lateral foot swelling degree was significant.Compared with the model group, Compound Qingfengteng Cataplasm could significantly improve the weight change of rats induced by CFA.Compound Qingfengteng Cataplasm reduced the secondary lateral foot swelling rats by injection of CFA.The level of IL-1βand TNF-α of AA rats in the model group were significantly higher than those in the blankcontrol group.AA rats had numerous inflammatory cell infiltration, pannus formation and severe articular cartilage damage in synovial tissue.The level of IL-1β and TNF-α in each dose group was significantly lower than those in the model group(P<0.05).The high-dose group had the most significant effect on reducing synovial lesions and articular cartilage injury in AA rats.Conclusion Compound Qingfengteng Cataplasm can obviously improve the performance of AA,and it may relate its therapeutic mechanism to the inhibition of the levels of related inflammatory factors.
