摘要
本研究应用DNA条形码技术,以ITS序列为目的片段对口岸截获的拟石莲属多肉植物进行快速鉴定。对11种36份拟石莲属供试样本ITS片段进行通用引物扩增,并双向测序拼接,利用MEGA7软件进行序列比对,分析各种类种内和种间K2P遗传距离,构建系统发育树。研究表明,供试材料ITS序列的种内差异显著小于种间差异,种内种间差异存在明显的间隔区,所有样品按种类在NJ树上严格聚成单系,种间区分明显。经初步判定,ITS具有作为拟石莲属多肉植物DNA条形码的潜在可能。
In this study,DNA barcode was used to quickly identify Echeveria succulents intercepted at the port by using ITS sequence as the target fragment.The ITS fragments of 36 samples from 11 species of Echeveria were amplified by universal primers and sequenced bi-directionally.Using the MEGA7 program,sequences were aligned,intra-and inter-specific K2P genetic distances were studied,and a phylogenetic tree was set up.The results showed that the intraspecific variation of ITS sequence was significantly smaller than that of interspecific variation,and thus created an obvious barcoding gap.All samples were clustered strictly into a monophyletic group in the NJ tree presenting an obvious inter-specific difference.After preliminary judgement,ITS had the potential to serve as a DNA barcode for Echeveria plants.
