叶用莴苣NAC基因家族的鉴定与热胁迫表达分析 被引量:1

Identification of NAC Gene Family in Leaf Lettuce and Their Expression under Heat Stress

摘要 NAC基因家族是植物特有的一类转录因子家族,在植物响应高温等非生物逆境胁迫中发挥重要作用。本研究通过对叶用莴苣基因组数据库进行查询鉴定,共有99个LsNAC基因,编码NAC蛋白161个。LsNAC基因在1号到9号染色体上均有分布,2个LsNAC基因未能定位在染色体上,其中6号染色体上编码的基因最多,8号染色体上编码的基因最少。叶用莴苣NAC蛋白有着相似的motif组成,分析表明,NAC总共有9个保守的motif,前8个motif分布在保守的N端,只有第9个motif位于序列不保守的C端,而且第9个motif仅存在于9个Ls NAC蛋白中。LsNAC基因的重复性鉴定分析表明,7对为片段重复基因,6对为串联重复基因。系统进化分析表明Ls NACs可以分为6个亚家族,通过对叶用莴苣热胁迫下的转录组数据进行分析,鉴定出13个对热胁迫响应的LsNAC基因,这些LsNAC基因在除第六亚家族之外,在其他五个家族中均有分布,其中第三个亚家族含有的热响应基因最多(4个LsNAC基因),RT-qPCR实时荧光定量分析进一步验证表明10个NAC基因在耐热材料中上调表达,3个下调表达。本研究为今后LsNAC基因在叶用莴苣中的耐热机理研究和相关抗逆育种提供了重要的理论基础。 The NAC gene were special TFs in plant and played an important role in response to heat stress and other abotic stress. In this study, 99 LsNAC genes and 161 NAC proteins were identified by searching and identifying the genome database of lettuce. LsNAC genes were distributed on chromosomes 1 to 9, two LsNAC gene could not be located on chromosome, of which the most genes were encoded on chromosome 6 and the least on chromosome 8. The NAC protein of leaf lettuce has a similar motif composition, the analysis showed that NAC has a total of nine conservative motif, and the eight motif were distributed on the conservative N-terminal, only the 9 thmotif was located in the C-terminal which was not conserved in the sequence, and the 9 th motif only existed in 9 LsNAC proteins. The repeatability analysis of LsNAC gene showed that 7 pairs were fragment repeat genes and 6 pairs were tandem repeat genes. Phylogenetic analysis showed that it could be divided into 6 subfamilies. By analyzing the transcriptome data of leaf lettuce under heat stress, 13 NAC genes in response to heat stress were identified. It was distributed in the other five families except for the sixth subfamily, of which the third subfamily contains the most genes in response to heat stress(4 LsNAC genes). RT-qPCR analysis further confirmed that 10 NAC genes were up-regulated and 3 down-regulated in heat-resistant materials. This study provides an important theoretical basis for the study of heat-resistant mechanism of LsNAC gene in leaf lettuce and related stress-resistant breeding in the future.
