摘要
目的:建立一测多评法同时测定中药苍耳子生品和炮制品中5个酚酸类成分含量的方法。方法:采用ODS HYPERSIL C_(18)色谱柱(4.6 mm×250 mm,5μm),检测波长为254 nm,柱温为30℃,流速为1.0 mL·min^(-1),进样量为10μL,流动相为0.1%磷酸水溶液(A)-甲醇(B),梯度洗脱。以绿原酸为内参物,建立绿原酸与咖啡酸、异绿原酸A、洋蓟素、异绿原酸C之间的相对校正因子,同时采用一测多评法和常规外标法测定中药苍耳子生品和炮制品中5个酚酸类成分的含量,对2种方法的含量测定结果进行比较。结果:一测多评法与外标法的含量测定结果无显著性差异,所建立的相对校正因子的耐用性良好。结论:一测多评法用于中药苍耳子生品和炮制品中酚酸类成分的含量测定具有可行性及科学性,可用于中药苍耳子的质量控制。
Objective:To establish a method for the simultaneous determination of five phenolic acids in the traditional Chinese medicine Xanthii Fructus with the quantitative analysis of multi-components by single marker(QAMS).Methods:The analysis was performed on an HYPERSIL C_(18)column(4.6 mm×250 mm,5μm),the detection wavelength was 254 nm,the column temperature was 30℃,the flow rate was 1.0 mL·min^(-1),the injection volume was 10μL,the mobile phase was 0.1%phosphoric acid(A)-methanol(B)with gradient elution.Using chlorogenic acid as the internal reference substance,the relative correction factors(RCFs)of caffeic acid,isochlorogenic acid A,cynarin and isochlorogenic acid C to chlorogenic acid were determined.At the same time,QAMS and the conventional external standard method were adopted to determine the contents of five phenolic acids in the Xanthii Fructus,and the results of the two methods were compared.Results:There was no significant difference between the content determination results of QAMS and the external standard method,and the established relative correction factors(RCFs)had good durability.Conclusion:QAMS is feasible and scientific for the determination of phenolic acids in the traditional Chinese medicine Xanthii Fructus.It can be used for the quality control of the traditional Chinese medicine Xanthii Fructus.
