刺五加白茯片的遗传毒性评价 被引量:1

Genotoxicity Evaluation of Acanthopanax Senticosus White Poria Cocos Tablets

摘要 目的评价刺五加白茯片的遗传毒性。方法本研究根据《保健食品检验与评价技术规范》(2003年版)采用Ames试验:设8μg/皿、40μg/皿、200μg/皿、1000μg/皿和5000μg/皿5个剂量组及溶剂对照组(蒸馏水)、自发对照组和阳性对照组,在加和不加S9的试验条件下进行平板掺入法试验。每组作3个平行皿,重复试验一次,测定各组回变菌落数。骨髓细胞微核试验:受试物设2500 mg/kg,5000 mg/kg和10000 mg/kg 3个剂量组,阳性对照为40 mg/kg环磷酰胺。采用30 h两次给药法,两次灌胃间隔24 h,于第二次给受试物后6 h脱颈椎处死动物,取胸骨髓进行制片,固定,Giemsa染色后,在油镜下计数1000个嗜多染红细胞(PCE)中含微核细胞数,计算微核细胞率。观察200个红细胞中嗜多染红细胞与成熟红细胞的比值(PCE/NCE)。精子畸形试验:受试物设2500 mg/kg,5000 mg/kg和10000 mg/kg 3个剂量组,阳性对照为40 mg/kg环磷酰胺。连续灌胃5 d,于首次给受试物后第35 d,脱颈椎处死动物取双侧附睾进行制片,甲醇固定,1%伊红染色后,在高倍镜下每只动物计数1000条完整精子,记录精子畸形、畸形类型及计算精子畸形率。3项试验综合评价刺五加白茯片的遗传毒性。Ames试验的单因素方差分析、骨髓细胞微核试验的卡方检验、精子畸形试验的秩和检验,上述数据统计均用SPSS 21.0软件进行处理,以P<0.05为差异有统计学意义。结果在Ames试验中,刺五加白茯片不引起菌株突变;在小鼠骨髓嗜多染红细胞(PCE)微核试验中,刺五加白茯片不增加微核率;在小鼠精子畸形试验中,刺五加白茯片不增加精子畸形率。结论在当前实验条件下,刺五加白茯片不具备遗传毒性。 Objective To evaluate the genetic toxicity of Acanthopanax senticosus white Poria Cocos tablets.Methods Based on the criterion of testing and evaluating health foods(2003 edition)-Ames test,five dose groups of 8,40,200,1000 and 5000/plate,solvent control group(distilled in water),spontaneous control group and positive control group were established.Plate incorporation test was conducted with or without S9.Three parallel plates were made for each group,and the test was repeated once.The number of revertant colonies in each group was measured.Micronucleusassay of bone marrow cells was performed.Three dose groups(2500,5000,10000 mg/kg)were used and the positive control was 40 mg/kg CP.The method of 30 h double administration was adopted,and the interval between two intragastric administrations was 24 h.The animals were killed by cervical vertebrae removal after 6 hours of the second administration.After Giemsa staining,the number of micronucleus cells in 1000 PCE were counted under oil microscope,and the micronucleus cell rate(‰)was calculated.The ratio of polychromatic erythrocytes to mature PCE/NCE in 200 erythrocytes were observed.Sperm abnormality test was conducted.Three dose groups(2500,5000,and 10000 mg/kg)were established and the positive control was 40 mg/kg CP.On the 35 th day after the first administration of the test substance,the animals were killed by removing the cervical vertebrae,and bilateral epididymis were taken for sectioning.After fixed with methanol and stained with 1%eosin,1000 intact spermatozoa were counted in each animal under high power microscope.One-way ANOVA,chisquare tests and rank sum tests were performed by using SPSS 21.0 software,and P<0.05 was considered as statistically significant.Results Acanthopanax senticosus white Poria Cocos tablets did not increase mutation in Ames test,not promote micronucleus rate in micronucleus test of bone marrow cells,and did not add the rate of sperm abnormality.Conclusion Acanthopanax senticosus white Poria Cocos tablets had no genetic toxicity under the current experimental condition.
