摘要
为制备抗蒜氨酸抗体,探讨其应用价值,将蒜氨酸与卵清蛋白结合,加入弗氏佐剂,常规免疫家兔;用间接血凝试验、间接免疫荧光试验检测蒜氨酸抗体及其效价;利用蒜氨酸抗体和蒜氨酸荧光抗体探讨蒜氨酸在组织细胞的分布特点及蒜氨酸的抗菌作用和可能机制,探索蒜氨酸抗体的潜在应用价值。结果显示,免疫家兔抗蒜氨酸抗体检测鉴定结果均为阳性,抗体效价达到1∶64;抗体能够准确反映蒜氨酸在组织细胞的分布特点,初步显示蒜氨酸的抑菌作用。蒜氨酸通过与异种蛋白偶联形成完全抗原,免疫家兔获得抗蒜氨酸抗体,并可将其应用于基础与临床研究。
To produce anti-alliin antibody and explore its application value, alliin was used to immunize rabbits after being mixed with egg albumin and Freund’s adjuvant. The anti-alliin antibody and its titer were detected by indirect agglutination test and indirect immunofluorescence test. Anti-alliin antibody and anti-alliin fluorescent antibody were used to determine alliin tissue and cellular distribution characteristics, as well as its antibacterial mechanism. The results showed that the antibody titer reached 1:64(all the outcomes were positive), and the alliin tissue distribution characteristics could be correctly reflected, and the antibacterial mechanism of alliin was preliminarily demonstrated. In conclusion, alliin can form complete antigens by coupling with heterologous proteins, which can be used to make anti-alliin antibody by rabbit immunization, and the antibody may be applied to basic and clinical researches.
