杨梅素抗抑郁作用及对神经递质的调控机制 被引量:3

摘要 目的探究杨梅素抗抑郁作用及对神经递质的调控机制。方法50只小鼠随机分空白对照组,模型组,杨梅素高、低剂量组(50、25 mg/kg),氟西汀组(10 mg/kg),采用慢性不可预知性刺激方法构建抑郁模型,造模同时灌胃给药,空白对照组、模型组则灌胃给予生理盐水。采用旷场实验、悬尾实验、强迫游泳实验检测各组小鼠自主活动量、悬尾不动时间、游泳不动时间,HPLC法测定小鼠脑中5-HT、NE及DA等单胺递质的水平,荧光分光光度法检测单胺氧化酶(MAO)活性。结果与模型组比较,给药组小鼠水平垂直活动量约是模型组的2倍,悬尾和游泳不动时间约为60~80 s,缩短了50~100 s,差异具有统计学意义(P<0.05)。与模型组比较,杨梅素高剂量组小鼠脑中5-HT、NE及DA单胺递质水平升高(P<0.05),且与氟西汀组相当,杨梅素高剂量组小鼠大脑MAO-A和MAO-B活性均降低(P<0.05),小鼠抑郁行为得到改善。结论梅素能预防并改善慢性不可预知应激诱导的抑郁样行为,其机制可能是抑制MAO活性,增加大脑中枢神经递质5-HT、NE及DA水平。
