摘要
目的:观察加味益气通玄方对大鼠脑缺血再灌注损伤(CRI)的作用机制。方法:48只改良的Zea Longa线栓法造模,高、中、低剂量组分别给予27.68g/(kg·d)、13.84g/(kg·d)、6.92g/(kg·d)加味益气通玄方,阳性药物组用60mg/(kg·d)阿司匹林治疗14 d。Zea Longa5分法、氯化三苯基四氮唑(TTC)染色评估加味益气通玄方对大鼠CRI的保护作用。Western blot检测内质网应激介导凋亡通路相关蛋白GRP78、CHOP、caspase12的表达。结果:加味益气通玄方能明显降低大鼠的神经行为学评分、减少梗塞体积,降低GRP78、CHOP、caspase12的表达。结论:加味益气通玄方改善大鼠CRI的机制可能与抑制内质网应激有关。
Objective:To observe the mechanism of Modified Yiqi Tongxuan Recipe on cerebral ischemia reperfusion injury(CRI) in rats. Methods: Forty-eight modified Zea Longa sutures were used to create models. The high, medium and low dose groups were given 27.68 g/(kg·d), 13.84 g/(kg·d), 6.92 g/(kg·d) Modified Yiqi Tongxuan Recipe, respectively. The positive drug group were treated with 60 mg/(kg·d) aspirin for 14 days. Zea Longa 5-point method and triphenyltetrazolium chloride(TTC) staining were used to evaluate the protective effect of Modified Yiqi Tongxuan Recipe on CRI in rats. Western blot was used to detect the expression of GRP78, CHOP, and caspase12 related to the apoptosis pathway mediated by endoplasmic reticulum stress. Results: Modified Yiqi Tongxuan Recipe can significantly reduce the neurobehavioral score of rats, reduce infarct volume, and reduce the expression of GRP78, CHOP, and caspase12. Conclusion: The mechanism of Modified Yiqi Tongxuan Recipe in improving CRI of rats may be related to the inhibition of endoplasmic reticulum stress.
