摘要
目的探讨中国北方汉族男性组蛋白去乙酰化酶2(histone deacetylase 2,HDAC2)基因多态性与酒精使用情况对认知功能的影响。方法纳入中国北方地区的汉族男性504名,采用酒精使用障碍筛查量表(alcohol use disorders identification test,AUDIT)进行筛查并分组为高风险饮酒组(n=194)、低风险饮酒组(n=186)和对照组(n=124)。采用简易视觉图形记忆测验(brief visuospatial memory test-revised,BVMT-R)、数字符号替换测验(digit symbol substitution test,DSST)、数字广度测试(digit span test,DST)评估3组人群认知功能情况。提取受试者外周血中DNA,采用Agena MassARRAY SNP基因分型法检测HDAC2的rs9481408、rs6568819、rs9488289基因多态性。结果与高风险饮酒组及对照组比较,低风险饮酒组的DSST得分较高(P<0.05)。根据各位点基因型分层分析,结果显示对于rs9481408、rs6568819、rs9488289位点,均为携带CC基因型者中低风险饮酒组DSST得分高于对照组,差异具有统计学意义(P<0.05)。结论中国北方汉族男性人群HDAC2基因rs9481408、rs6568819、rs9488289位点的CC基因型与低风险饮酒者部分认知功能较好可能存在相关性。
Objective To explore the effect of alcohol use and histone deacetylase 2(HDAC2)gene polymorphisms on cognitive function of Male Han Population in Chinese Northern Han Population.Methods The high-risk drinking group(n=194)was screened out according to the Alcohol Use Disorders Identification Test(AUDIT),and the rest of people was divided into two groups:low-risk drinking group(n=186)and control group(n=124)according to whether ever drinking.The Brief Visuospatial Memory Test-Revised(BVMT-R),the Digit Symbol Substitution Test(DSST)and Digit Span Test(DST)were used to measure their cognitive function.The DNA extracted from the peripheral blood were analyzed to determine the HDAC2 rs9481408,rs6568819 and rs9488289 gene polymorphism by Agena MassARRAY.Results The scores of DSST were significantly higher in low-risk group than in either high-risk group or control group(P<0.05).Further analysis showed that the DSST scores of the HDAC2 CC gene carriers were significantly higher in the low-risk drinking group than in the control group(P<0.05).Conclusion HDAC2 CC genotype of rs9481408,rs6568819 and rs9488289 may be associated with cognitive functions in some cognitive domain among moderate drinkers.
