异丙肾上腺素通过诱导心肌线粒体氧化应激在小鼠心律失常中的作用 被引量:2

摘要 目的探讨异丙肾上腺素(ISO)通过诱导心肌线粒体氧化应激诱发小鼠心律失常的机制。方法小鼠腹腔注射异丙肾上腺素前后分别记录心电图。提取小鼠心肌组织,检测线粒体呼吸链复合物I、ATP、ROS的变化水平。光标测技术检测心脏动作电位的变化情况。结果注射ISO后,小鼠的心率明显增加,起搏诱发心律失常的机率增加。ATP含量和线粒体呼吸链复合物I浓度降低,ROS含量增加。光标测结果显示心肌动作电位时程缩短。结论异丙肾上腺素通过影响心肌线粒体氧化应激诱导心律失常的发生。 Objective To study the mechanism of isoproterenol triggering arrhythmia through myocardial mitochondrial oxidative stress in mice.Methods An electrocardiogram was recorded before and after intraperitoneal injection of isoproterenol.Myocardial tissues were extracted from the mice to measure the changes in the levels of mitochondrial respiratory chain complex I,adenosine triphosphate(ATP),and reactive oxygen species(ROS).The cardiac action potential was measured by optical mapping.Results After isoproterenol injection,the mice had a significantly increased heart rate,significantly more frequent arrhythmia triggered by pacing,significantly decreased levels of ATP and mitochondrial respiratory chain complex I,a significantly increased ROS level,and a significantly shortened cardiac action potential duration.Conclusion Isoproterenol induces oxidative stress in myocardial mitochondria to trigger arrhythmia.
