摘要
为明确山东乳山迟采酿酒葡萄‘贵人香’引发贵腐作用的菌种及培养条件,以形态学为基础,结合生理生化及分子生物学方法对被侵染的‘贵人香’葡萄进行菌种分离鉴定。结果表明,从被侵染的葡萄中筛选出68株菌种,通过形态学观察、生理生化鉴定以及ITS rDNA序列分析,确定有3株贵腐菌(Botrytis cinerea),分别命名为GRX06、GRX08、GRX62。各菌株均在改良PDA培养基上生长良好,但GRX06以葡萄糖为碳源、牛肉膏为氮源的培养基上表现较好,GRX08以甘露醇为碳源、硝酸钾为氮源的培养基上生长较好,GRX62以甘露醇为碳源、酵母粉为氮源的培养基上表现较佳。GRX06和GRX08以改良PDA培养基在pH 6和25℃条件下生长良好,GRX62以改良PDA培养基在pH 8和20℃下生长良好,光照时间对这些菌株的生长无显著影响。
In order to clarify the strains causing noble rot on late harvested'Italian Riesling'in Rushan region of Shandong and to optimize the culture conditions,the strains from botrytized'Italian Riesling'grape were isolated and identified on the basis of morphology,combined with physiological,biochemical and molecular biological methods.The results showed that 68 strains were isolated from infected grapes and three strains of Botrytis cinerea were identified through morphological observation,physiological and biochemical identification and the ITS rDNA sequence analysis,named,GRX06,GRX08,GRX62,respectively.Each strain grew well on modified PDA medium.But GRX06 grew better on medium with glucose as carbon source and beef paste as nitrogen source,GRX08 grew better on medium with mannitol as carbon source and potassium nitrate as nitrogen source,while GRX62 grew better with mannitol as carbon source and yeast powder as nitrogen source.GRX06 and GRX08 grew well under 25℃with pH 6 of PDA,while GRX62 grew well under pH 8 of PDA and 20℃.Light application time had no significant effect on of these strains.
