松果菊苷对葡聚糖硫酸钠诱导的实验性结肠炎模型大鼠的保护作用 被引量:2

摘要 目的:探讨松果菊苷(ECH)对葡聚糖硫酸钠(DSS)诱导的实验性结肠炎大鼠模型炎症的调节作用,并初步分析其相关作用机制。方法:将SD大鼠随机分为对照组、DSS组、DSS+低剂量ECH(ECH-L)组和DSS+高剂量ECH(ECH-H)组,每组10只大鼠。DSS组、DSS+ECH-L组和DSS+ECH-H组大鼠连续7 d给予饮用4%DSS,然后正常饮水3 d;其中DSS+ECH-L组和DSS+ECH-H组分别每天给予20和50 mg/kg ECH灌胃,DSS组给予等体积生理盐水,对照组大鼠在整个实验过程中正常饮水。各组大鼠分别进行疾病活动指数(DAI)评分。收集各组大鼠结肠并测量长度;HE染色观察各组大鼠结肠组织病理学表现;免疫组化染色观察各组大鼠结肠组织中白细胞介素6(IL-6)和肿瘤坏死因子α(TNF-α)表达情况;ELISA法测定各组大鼠结肠组织中IL-1β、IL-6、IL-10和TNF-α水平;免疫荧光染色观察各组大鼠结肠组织中Toll样受体2(TLR2)表达情况;用Western blot法检测各组大鼠结肠组织中TLR2、TLR4和核因子κB(NF-κB)的表达水平。结果:与对照组比较,DSS组大鼠的DAI评分显著升高(P<0.01),结肠长度显著缩短(P<0.01),结肠组织的病理形态结构出现明显破坏,结肠组织中IL-1β、IL-6、TNF-α、TLR2、TLR4和NF-κB水平均显著增高(P<0.01),IL-10水平显著降低(P<0.01)。与DSS组比较,DSS+ECH-L和DSS+ECH-H组中上述指标均得到显著改善(P<0.05或P<0.01),其中DSS+ECH-H组效果尤为明显(P<0.01)。结论:ECH能减轻DSS诱导的实验性结肠炎大鼠的症状,降低炎症反应,且这些作用可能与其降低TLRs/NF-κB信号通路活性有关。 AIM:To investigate the effect of echinacoside(ECH) on inflammatory regulation in an experimental colitis rat model induced by dextran sodium sulfate(DSS),and to preliminarily analyze its mechanism.METHODS:SD rats were randomly divided into control group,DSS group,DSS+low-dose ECH(ECH-L) group and DSS+high-dose ECH(ECH-H) group,with 10 rats in each group.The rats in DSS group,DSS+ECH-L group and DSS+ECh-H group were given 4% DSS for 7 d,and then normal water for 3 d.Moreover,during the period,the rats in DSS+ECH-L group and DSS+ECh-H group were given 20 and 50 mg/kg ECH gavage daily,respectively,and the rats in DSS group were given equal volume of normal saline.The rats in control group drank normal water throughout the experiment.The disease activity index(DAI) was scored.The colon tissues of the rats in each group were collected and their length was measured.The histopathological images of the colon were observed by HE staining.The expression of interleukin-6(IL-6) and tumor necrosis factor-α(TNF-α) in colon tissues of the rats was observed by immunohistochemical staining.The levels of IL-1β,IL-6,IL-10 and TNF-α in colon tissues of the rats were measured by ELISA.The expression of Toll-like receptor 2(TLR2) in colon tissues was observed by immunofluorescence staining.The expression levels of TLR2,TLR4 and nuclear factor-κB(NF-κB) in colon tissues of the rats in each group were determined by Western blot.RESULTS:Compared with control group,the DAI score in DSS group was significantly increased(P<0.01),the length of the colon was significantly shortened(P<0.01),the pathological structure of colon tissue was significantly damaged,and the levels of IL-1β,IL-6,TNF-α,TLR2,TLR4 and NF-κB in colon tissues were significantly increased(P<0.01),while the level of IL-10 was significantly decreased(P<0.01).Compared with DSS group,the above indexes were significantly improved in DSS+ECH-L group and DSS+ECH-H group(P<0.05 or P<0.01),and the effect was particularly significant in DSS+ECH-H group(P<0.01).CONCLUSION:ECH attenuates the symptoms and reduces the inflammatory response of experimental colitis in rats induced by DSS,and these effects may be related to the decrease in the activity of TLRs/NF-κB pathways.
