不同剩余采食量下滩羊能量代谢分析 被引量:3

Energy Metabolism Analysis of Tan Sheep with Different Residual Feed Intake

摘要 本研究旨在揭示不同剩余采食量(RFI)下滩羊血浆内源性能量代谢物的变化,筛选出对滩羊RFI有影响作用的差异代谢物。筛选极高、极低RFI滩羊各16只并采集血液,应用代谢组学技术对血浆样品进行检测,结合主成分分析(PCA)和正交偏最小二乘判别分析(OPLS-DA),以变异倍数(FC)>1.30或<0.77,变量投影重要度(VIP)>1和P<0.05为标准筛选差异代谢物。结果显示:极低RFI公羊vs极高RFI公羊组中D-木酮糖-5-磷酸、L-鸟氨酸盐酸盐下调;极低RFI母羊vs极高RFI母羊组中腺嘌呤、柠檬酸、L-天冬氨酸上调,L-瓜氨酸下调;极低RFI羊vs极高RFI羊组中腺嘌呤、柠檬酸上调,L-苏氨酸、D-木酮糖-5-磷酸下调。经受试者工作特征(ROC)曲线分析发现极低RFI母羊vs极高RFI母羊组中柠檬酸的ROC曲线下面积(AUC)>0.9。综上可知,性别会导致滩羊能量代谢不同,柠檬酸有望成为影响母滩羊RFI的候选生物标志物。 The aim of this study was to reveal the changes of endogenous energy metabolites in plasma of Tan sheep under different residual feed intake(RFI),and to screen out the differential metabolites that have an effect on RFI of Tan sheep.Blood samples were collected from 16 Tan sheep with very high RFI and very low RFI,respectively,metabolomics was used to detect plasma samples.Combined with principal component analysis(PCA)and orthogonal partial least squares discriminant analysis(OPLS-DA),differential metabolites were screened according to the criteria of multiple of variation(FC)>1.3 or<0.77,variable projection importance(VIP)>1 and P<0.05.The results showed that D-xylose-5-phosphate and L-ornithine hydrochloride were down regulated in very low RFI rams vs very high RFI rams group;adenine,citric acid and L-aspartate were up-regulated and L-citrulline was down regulated in very low RFI ewes vs very high RFI ewes group;adenine and citric acid were up-regulated and L-threonine and D-xylose-5-phosphate were down regulated in very low RFI sheep vs very high RFI sheep group.The receiver operating characteristic(ROC)curve analysis showed that the area under the ROC curve(AUC)of citric acid in very low RFI vs very high RFI ewe group was more than 0.9.In conclusion,gender can lead to different energy metabolism of Tan sheep,and citric acid is expected to be a candidate biomarker for influencing RFI of female Tan sheep.
