摘要
研究组织中蛋白激酶Cε(PKCε)表达对乳腺癌妇女临床病理特点的影响。方法免疫组化法进行检测。收集的新鲜乳腺癌组织,使用4%福尔马林固定14 h,石蜡包埋,切成5µm的薄片。脱蜡,水化。山羊血清封闭,降低非特异性染色的概率。将多余血清倒出,加入一抗PKCε(1∶85稀释),冰箱4℃过夜,室温下放置20 min,缓冲液5 min/次,共3次。加入二抗,缓冲液洗3次(3 min/次);显色3 min,流水冲洗10 min,复染,脱水封片。将乳腺癌阳性切片作为阳性对照,鼠免疫球蛋白G(IgG)及磷酸盐缓冲液(PBS)代替一抗作为阴性对照。结果PKCε在56例非特殊型乳腺癌中有51例表达为阳性,阳性率达到91.07%。在38例导管原位癌中有31例为阳性表达,阳性率为81.58%。113例乳腺癌患者的切除组织中,不同年龄段之间存在较大差异,大部分集中在47~75的年龄范围内。在乳腺癌肿瘤直径中,直径>5 cm的患者中有93.33%的PKCε具有高表达。在TNM分期中,Ⅲ~Ⅳ期患者34例乳腺癌组织中,31例PKCε具有高表达,阳性率达到91.18%。结论组织中蛋白激酶Cε的表达升高与乳腺癌的患者的年龄、肿瘤直径、TNM分期及淋巴结转移之间具有相关性,PKCε的表达水平能够成为检测乳腺癌的重要指标。
【Objective】To study the effect of protein kinase Cεexpression in tissues on clinicopathological characteristics of breast cancer women.【Methods】Immunohistochemical method was used for detection.The collected fresh breast cancer tissues were fixed with 4%formalin for 14 hours,embedded in paraffin,and cut into 5μm slices.After dewaxing and hydration,normal goat serum was used for blocking to reduce the probability of non-specific staining.The excess serum was poured out,the primary antibody PKCε(diluted 1:85)was added,it was kept in a refrigerator at 4℃ overnight,placed at room temperature for 20 minutes,and washed with buffer solution 5 minutes/time for a total of 3 times.Secondary antibody was added,and it was washed with buffer solution 3 times(3min/time);coloration time was 3 minutes,it was rinsed with running water for 10 minutes,then counterstain,dehydrate and mount.The breast cancer positive section was used as a positive control,and mouse IgG and PBS were used as a negative control instead of the primary antibody.【Results】PKCεwas positive in 51 of 56 cases of non-specific breast cancer,and the positive rate reached 91.07%.In 38 cases of ductal carcinoma in situ,31 cases were positive,and the positive rate was 81.58%.Among the removed tissues of 113 breast cancer patients,there were large differences between different age groups,most of which were concentrated in the age range of 47-75.Among the diameters of breast cancer tumors,93.33%of patients with a diameter greater than 5 cm had high expression of PKCε.In TNM staging,among 34 breast cancer tissues of patients with stage III-IV,31 cases of PKCεhave high expression,with a positive rate of 91.18%.【Conclusion】The elevated expression of protein kinase Cεin tissues is correlated with the age,tumor diameter,TNM stage and lymph node metastasis of breast cancer patients.The expression level of PKCεcan be an important indicator for detecting breast cancer。
