摘要
本研究旨在分析头孢噻呋钠在牛体内的残留消除规律,进而为休药期的制定提供依据。20头健康试验牛按体质量以2.2 mg/kg肌内注射头孢噻呋钠,连续给药3 d,分别在最后1次给药后0.5,3,5,7,9 d宰杀所有试验牛并采集背最长肌、注射部位、肝、肾以及脂肪样品。样品用二硫赤藓糖醇提取后,经碘乙酰胺衍生化后调节pH至2.3,最后使用HLB固相萃取柱净化,氮吹复溶后用高效液相色谱串联质谱(LC-MS/MS)检测。结果显示:给药后0.5 d肾组织中的药物浓度最高,为3772.02μg/kg,且所有组织的药物浓度均低于最大残留限量(MRL)。参照EMEA制定的头孢噻呋钠在牛上的MRL,采用休药期计算软件WT1.4计算各组织的休药期,各组织的休药期分别为背最长肌0.00 h、注射部位92.01 h、肝31.77 h、肾68.13 h、脂肪25.74 h,计算可得注射部位的休药期最长,为92.01 h。结果提示,头孢噻呋钠在牛体内代谢迅速,消除较快,建议将休药期定为4 d。
The aim of this study was to analyze the residue elimination pattern of ceftiofur sodium in cattle and to provide a basis for the establishment of a rest period.Twenty healthy test cattle were injected intramuscularly at 2.2 mg/kg for 3 d,and all test cattle were slaughtered and samples were collected from the longest dorsal muscle,injection site,liver,kidney and fat on d 0.5,3,5,7 and 9 after the last dose.The samples were extracted with dithio-erythritol,derivatized by iodoacetamide and pH was adjusted to 2.3.The samples were cleaned up using an HLB solid phase extraction column,nitrogen blown and then detected by high performance liquid chromatography tandem mass spectrometry(LC-MS/MS).The results showed that the highest concentration of 3772.02μg/kg was found in renal tissues on d 0.5 after administration and all tissues were below the maximum residue limit(MRL)on d 0.5.Referring to the MRLs developed by EMEA for ceftiofur sodium in cattle,the resting period of each tissue was calculated using the resting period calculation software WT1.4.The resting periods of each tissue were 0 h for dorsal muscle,92.01 h for injection site,31.77 h for liver,68.13 h for kidney and 25.74 h for fat,and the resting period of 92.01 h for the injection site was the longest.The results suggest that ceftiofur sodium is rapidly metabolized and eliminated in cattle,and it is recommended that the rest period be set at 4 d.
