摘要
目的探索水通道蛋白9(AQP9)在慢传输型便秘(STC)大鼠结肠中的表达情况,及其作用机制。方法SD大鼠灌胃给予复方地芬诺酯混悬液建立大鼠慢传输型便秘(STC)模型,并随机分成模型组、对照组及实验组,每组12只;另取12只正常大鼠作为空白组。STC模型建立后,空白组和模型组均灌胃给予0.9%NaCl 4 mL;对照组灌胃给予0.13 mg·mL^(-1)莫沙必利混悬液4 mL;实验组灌胃给予0.15 g·mL^(-1)麻仁丸混悬液4 mL。4组大鼠每天给药1次,连续给药7 d。待末次给药结束后,进行碳末推进实验检测,用Western blot法检测4组结肠组织中AQP9的表达情况。结果实验组、对照组、模型组和空白组的碳末推进率分别为(70.18±2.84)%,(69.28±2.89)%,(51.56±3.41)%和(71.25±5.61)%,AQP9蛋白相对表达量分别为0.34±0.02,0.35±0.03,0.20±0.02和0.42±0.02。模型组的上述指标与其他3组比较,差异均有统计学意义(均P<0.01)。结论结肠中AQP9低表达水平会导致便秘。
Objective To explore the expression of aquaporin 9(AQP9)in the colon of the slow transit constipation(STC)rats and its mechanism.Methods SD rats were given compound diphenoxylate suspension by gavage to establish the model of STC,and were randomly divided into model,control and experimental groups with 12 rats per group.Another 12 normal rats were taken as the blank group.After the STC model was established,the blank and model groups were given 0.9%NaCl 4 mL by gavage;the control group was given 0.13 mg·mL^(-1) mosapride suspension 4 mL by gavage;the experimental group was given 0.15 g·mL^(-1) Maren pill suspension 4 mL by gavage.Four groups were administered once a day for 7 days.After the last administration,the carbon powder propulsion test was carried out,and the expression of AQP9 in the colon was detected by Western blot.Results The carbon powder propulsion rates of experimental,control,model and blank groups were(70.18±2.84)%,(69.28±2.89)%,(51.56±3.41)%and(71.25±5.61)%,the relative expression levels of AQP9 protein were 0.34±0.02,0.35±0.03,0.20±0.02 and 0.42±0.02.There were statistically significant between model group and other three groups(all P<0.01).Conclusion Low levels of AQP9 expression in the colon can cause constipation.
