摘要
目的探讨基质金属蛋白酶3(MMP3)在非小细胞肺癌(NSCLC)中的表达及对患者预后的影响。方法采用免疫组织化学(IHC)染色法检测MMP3在76份NSCLC组织及癌旁组织中的表达,并分析MMP3表达与组织病理分级、淋巴结转移、TNM分期、年龄及患者预后的关联性;利用基因表达综合(GEO)数据库分析MMP3 mRNA在NSCLC组织中的表达情况;利用基因表达谱表达分析(GEPIA)数据库分析MMP3 mRNA表达与患者预后的关系。结果与癌旁组织比较,MMP3 mRNA和蛋白在NSCLC组织中的表达均明显升高(P<0.05),且其表达水平与患者肿瘤直径、淋巴结转移和TNM分期呈明显正相关(P<0.05)。生存分析显示,MMP3阳性表达、MMP3 mRNA高表达与NSCLC患者的不良预后相关(P<0.05)。结论MMP3在NSCLC组织中高表达,可作为NSCLC治疗的潜在靶点和预后标志物。
Objective To explore the expression of matrix metalloproteinase 3(MMP3)in non-small cell lung cancer(NSCLC)and its effect on the prognosis of the patients.Methods The MMP3 protein expression in 76 samples of NSCLC tissue and paracancerous tissue was detected by immunohistochemistry(IHC),and the relationships between MMP3 expression with the histopathological grading,lymph node metastasis,TNM stage,age and the prognosis of the patients were analyzed.The expression of MMP3 mRNA in NSCLC tissues was analyzed by using the GEO database.The relationship between the MMP3 mRNA expression and the prognosis of NSCLC patients was analyzed by using the GEPIA database.Results Compared with paracancerous tissues,the expression of MMP3 mRNA and protein in NSCLC tissues was significantly increased(P<0.05),moreover its expression level was positively correlated with the tumor diameter,lymph node metastasis and TNM stage(P<0.05).The survival analysis showed that the positive expression of MMP3 protein and high expression of MMP3 mRNA were related to the poor prognosis of NSCLC patients(P<0.05).Conclusion MMP3 is highly expressed in NSCLC tissue,which can serve as a potential target and prognostic biomarker for NSCLC therapy.
