摘要
目的探讨桃仁红花煎对妊娠期糖尿病(GDM)大鼠及其子代结局的影响与机制。方法选择40只SPF级SD雌性大鼠、80只SPF级SD雄性大鼠。按照随机数字表法将40只雌性大鼠分为A、B、C、D组各10只,于动情期将雌性与雄性大鼠按1∶2合笼,用阴道涂片法确认雌性大鼠妊娠。妊娠第5天,B、C、D组在尾静脉注射链脲佐菌素(STZ)制备GDM模型,A组尾部注射等体积枸橼酸—磷酸氢二钠缓冲液。C组于建模当日用桃仁红花煎10 g/kg灌胃,D组用二甲双胍200 mg/kg灌胃,A、B组给予同体积生理盐水灌胃,每日1次,共给药19 d。分别于孕前及孕7、19 d用血糖仪检测空腹血糖(FPG),用酶联免疫吸附法检测胰岛素。孕19 d将大鼠麻醉后开腹取出胚胎并分离胚胎,计算各组胚胎存活数,称取胎鼠出生体质量。用HE染色法观察胎盘组织病理变化,用qRT-PCR检测胎盘组织中胰岛素样生长因子2 mRNA结合蛋白2(IGF2BP2)、核转录因子κB(NF-κB)mRNA表达,用Western blotting法检测胎盘组织IGF2BP2、NF-κB蛋白表达。结果孕7、19 d,B组FPG及胰岛素水平均高于A组(P均<0.05),C组上述指标均低于B组(P均<0.05)。与A组比较,B组胚胎存活数少、出生体质量低(P均<0.05);与B组比较,C组胚胎存活数多、出生体质量高(P均<0.05)。A组胎盘组织正常;B组胎盘组织分层模糊,细胞间隙增大,细胞布局紊乱;C、D组胎盘组织病理变化较B组轻。与A组比较,B组IGF2BP2及NF-κB mRNA、蛋白表达高(P均<0.05);与B组比较,C组上述指标表达低(P均<0.05)。C组与D组上述指标比较差异均无统计学意义(P均>0.05)。结论桃仁红花煎可降低GDM大鼠孕中血糖及胰岛素水平,提高子代胚胎成活率及体质量,改善胎盘组织,其机制可能与抑制IGF2BP2及NF-κB信号通路激活有关。
Objective To investigate the effects and mechanism of Taoren Honghua decoction on the gestational dia‑betes mellitus(GDM)rats and the outcomes of their offspring.Methods Forty SPF SD female rats and 80 SPF SD male rats were selected.Forty female rats were randomly divided into groups A,B,C and D,with 10 rats in each.During the estrous period,female and male rats were caged at the ratio of 1:2,and the pregnancy of female rats was confirmed by vagi‑nal smear method.On the 5th day of pregnancy,rats in the group B,group C and group D were injected with STZ in cau‑dal vein to prepare GDM model.Rats in the group A were injected with equal volume of citrate-disodium hydrogen phos‑phate buffer,rats in the group C were given Taoren Honghua decoction 10 g/kg intragastrically on the modeling day,rats in the group D were given metformin 200 mg/kg intragastrically;rats in the groups A and B were given the same volume of normal saline,once a day,for 19 days.Fasting blood glucose(FPG)and insulin were measured by ELISA before pregnan‑cy and on day 7 and 19 of pregnancy,respectively.After anesthesia on day 19 of gestation,the embryos were extracted and separated by laparotomy.The number of embryos in each group was calculated,and the amount of fetal body fat at birth was measured.The pathological changes of placental tissues were observed by HE staining,and the mRNA expres‑sion of insulin-like growth factor 2mRNA binding protein 2(IGF2BP2)and nuclear factorκB(NF-κB)in the placental tis‑sues were detected by qRT-PCR.The protein expression levels of IGF2BP2 and NF-κB in placental tissues were detected by Western blotting.Results On day 7 and 19 of pregnancy,the levels of FPG and insulin in the group B were higher than those in the group A(all P<0.05),and the above indexes in the group C were lower than those in group B(all P<0.05).Compared with the group A,group B had lower embryo survival and birth body fat(both P<0.05).Compared with the group B,group C had higher embryo survival and birth body fat(both P<0.05).The placental tissues of group A were normal.In the group B,the placental tissue layer was blurred,the intercellular space was enlarged,and the cell layout was disordered.The pathological changes of placenta tissues in the groups C and D were lighter than those in the group B.Compared with the group A,the mRNA and protein expression levels of IGF2BP2 and NF-κB in the group B were higher(all P<0.05).Compared with the group B,the expression of the above indexes in group C was lower(all P<0.05).There were no statistically significant differences in the above indexes between group C and group D(all P>0.05).Conclusions Taoren Honghua decoction can reduce blood glucose and insulin levels,increase the survival rate and body fat amount of offspring embryos,and improve placental tissue in GDM rats during pregnancy.The mecha‑nism may be related to inhibiting the activation of IGF2BP2 and NF-κB signaling pathways.
