铁心杉半同胞子代优株父本鉴定 被引量:3

Identification of paternal parents of half-sib progeny superior plants of the black-heart wood Chinese fir

摘要 【目的】组建铁心杉优良杂交组合,为铁心杉种质资源圃构建及种子园配置提供数据支撑和科学依据。【方法】选取铁心杉优良母树3576个半同胞子代中表现优良的幼苗优株以及403个候选父本、10个母本作为研究对象,筛选15个EST-SSR标记,采用ABI 370XL毛细管电泳检测PCR扩增产物,利用软件GeneMarker 2.20进行基因分型、软件PIC_CALC Version 0.6计算多态信息含量(PIC),GenALex6.5估算各个SSR位点的等位基因数目(NA)、期望杂合度(HE)、观测杂合度(HO);以等位基因分布频率为基础,CERVUS3.0.7软件和最大似然值法计算LOD值进行父本鉴定,计算NE-1P、NE-2P和NE-PP。【结果】共计选出50株铁心杉半同胞优株;15个SSR位点在所有样本中的平均等位基因数、平均观测杂合度和平均期望杂合度分别为9.867、0.475和0.679;15个SSR位点的多态信息含量PIC值在0.356~0.876,平均为0.634;平均NE-1P、NE-2P和NE-PP为0.872、0.802、0.702;50个半同胞子代在403个疑似父本中检测出的最似父本LOD值均大于0,其中TXS-14的繁殖贡献率最高为6%,TXS-20、TXS-201、TXS-204、TXS-399、TXS-5的繁殖贡献率次之,为4%,其余均为2%,共鉴定出42个真实父本,选出高配合力亲本杂交组合50对。【结论】已发表的SSR标记能够用来对铁心杉半同胞子代优株进行父本鉴定,研究结果将为铁心杉种质资源圃的构建提供理论依据和进一步开展铁心杉分子辅助杂交育种奠定基础。 【Objective】Form a good hybrid combination of the black-heart wood Chinese fir,and provide data support and scientific basis for the construction of the germplasm resource nursery and the configuration of the seed orchard.【Method】Select excellent seedlings from 3576 half-sib progeny of 10 excellent black-heart wood Chinese fir,as well as 403 candidate male parents and 10 female parents as the research objects.15 EST-SSR markers were screened and the PCR amplification products were detected using the automated capillary electrophoresis system ABI3730XL,the software GeneMarker2.20 was used for genotyping,PIC_CALC Version 0.6 was used to calculate the polymorphic information content(PIC),and GenALEx6.5 was used to estimate the number of alleles(NA)and expected heterozygosity(HE),Observed heterozygosity(HO);Based on allele distribution frequency,CERVUS 3.0.7 software and the maximum likelihood method calculate the LOD value for paternity identification,and calculate NE-1P,NE-2P and NE-PP.【Result】A total of 50 the black-heart wood Chinese fir semi-sib superior plants were selected,the average number of alleles,average observed heterozygosity and average expected heterozygosity of 15 SSR loci in all samples were 9.867,0.475 and 0.679 respectively.15 SSR loci The PIC value of polymorphic information content is 0.356-0.876,with an average of 0.634,the average NE-1P,NE-2P and NE-PP are 0.872,0.802,0.702.50 half-sibling offspring were detected in 403 suspected male parents,and the LOD valuesof the most similar paternal parent are all greater than 0,and the reproduction contribution rate of TXS-14 is the highest at 6%,followed by the reproduction contribution rate of TXS-20,TXS-201,TXS-204,TXS-399,and TXS-5(4%)and the rest is 2%.A total of 42 true male parents were identified,and 50 pairs of high combining ability parent hybrid combinations were selected.【Conclusion】The published SSR markers can be used to identify the paternity of the black-heart wood superior semi-sib progeny plants.The research results will provide a theoretical basis for the construction of the black-heart wood Chinese fir germplasm resource nursery and lay a foundation for further molecular-assisted hybrid breeding of the black-heart wood Chinese fir.
