miR-612通过调控TMPRSS4表达抑制IL-17对胃癌细胞增殖、迁移、侵袭的机制 被引量:1

摘要 目的探讨微小RNA-612(miR-612)对胃癌细胞增殖、迁移、侵袭的作用及其分子机制。方法qRT-PCR与Western印迹法检测白细胞介素(IL)-17对胃癌MKN-45细胞中miR-612、Ⅱ型跨膜丝氨酸蛋白酶(TMPRSS)4表达的影响;MTT与Transwell法检测IL-17对MKN-45细胞增殖、迁移及侵袭的影响。MKN-45细胞转染anti-miR-612或pcDNA-TMPRSS4后,分别检测细胞增殖、迁移及侵袭。Targetscan软件预测miR-612的靶基因,双荧光素酶报告基因验证靶基因。miR-612过表达验证MKN-45细胞增殖、迁移及侵袭。Western印迹法检测细胞内细胞周期蛋白(Cyclin)D1、基质金属蛋白酶(MMP)-2、MMP-9、p21蛋白的表达。结果IL-17可促进MKN-45细胞增殖、迁移及侵袭,促进TMPRSS4、CyclinD1、MMP-2、MMP-9表达,抑制miR-612、p21表达;转染anti-miR-612或pcDNA-TMPRSS4能够显著增加细胞的增殖、迁移及侵袭;miR-612可负性调控TMPRSS4;miR-612过表达可显著抑制由IL-17介导的胃癌细胞增殖、迁移及侵袭。结论miR-612通过抑制TMPRSS4表达而抑制IL-17对胃癌细胞增殖、迁移及侵袭的促进作用,其作用机制与CyclinD1、MMP-2、MMP-9表达下调及p21表达上调有关。
