响应面法优化红景天苷提取工艺研究 被引量:3

Study on the optimization of the extraction technology of salidroside by response surface methodology

摘要 以红景天为原料,分别考察超声提取法、乙醇回流提取法和乙醇浸提法对红景天苷含量的影响,以乙醇体积分数、浸提温度、超声时间以及料液比为考察因素,以红景天苷含量为考察指标,采用单因素试验结合响应面法分析与优化红景天苷提取条件。结果表明:提取效果为超声提取法(1.71±0.09)mg/g>乙醇浸提法(1.51±0.24)mg/g>乙醇回流提取法(1.47±0.05)mg/g,因此确定采用超声提取法结合乙醇浸提法提取红景天苷。乙醇体积分数60%、浸提温度30℃、超声时间2 h、料液比1∶80(g/mL)为最佳提取条件,在此条件下,提取液中红景天苷含量为2.35 mg/g。 Taking Rhodiola as raw material,the effects of ultrasonic extraction,ethanol reflux extraction and ethanol extraction on the content of salidroside were investigated respectively.Taking ethanol volume fraction,extraction temperature,ultrasonic time and solid-liquid ratio as investigation factors and salidroside content as investigation index,the extraction conditions of salidroside were analyzed and optimized by single factor test combined with response surface methodology.The results showed that the extraction effect was ultrasonic extraction(1.71±0.09)mg/g>ethanol extraction(1.51±0.24)mg/g>ethanol reflux extraction(1.47±0.05)mg/g.Therefore,ultrasonic extraction combined with ethanol extraction was selected to the extraction of salidroside.The optimum extraction condition was the ethanol volume fraction 60%,the extraction temperature 30℃,the ultrasonic time 2 h and the ratio of solid to liquid 1∶80(g/mL),under this condition,the content of salidroside in the extraction solution was 2.35 mg/g.
