2019—2020年河北省鸡细小病毒分子流行病学调查及NS基因序列分析 被引量:1

Molecular Epidemiological Investigation and NS Gene Sequence Analysis of Chicken Parvovirus in Hebei Province

摘要 为了解河北省鸡细小病毒(Chicken parvovirus,ChPV)在小肠有出血斑病变的鸡群中的流行及遗传变异情况,研究针对ChPV的非结构(NS)蛋白基因设计特异性检测引物,采用PCR方法对送检的151份样品进行检测,并对部分阳性病料中ChPV NS基因进行测序和遗传进化分析。流行病学调查结果显示:ChPV阳性率为48.3%;肉鸡和蛋鸡的阳性率分别为64.3%和44.7%,且幼龄鸡群的阳性率较高;4~5月份和7~8月份样品中ChPV的阳性率较高。序列分析显示测序病料中ChPV/CK/CH/HBSJZ/61株、ChPV/CK/CH/HBBD/62株和ChPV/CK/CH/HBSJZ/68株均与参考毒株ChPV/Poland/G090/2011核苷酸序列同源性最高(98.8%~99.1%),属于火鸡细小病毒(Turkey parvovirus,TuPV)亚群;ChPV/CK/CH/HBZJK/17株与参考毒株GX-CH-PV-17和GX-CH-PV-14的核苷酸序列同源性最高(均为99.7%),属于ChPV亚群。结果表明:河北省有出血斑病变的鸡小肠中ChPV检出率较高,测序样品中ChPV分别属于ChPV亚群和TuPV亚群。 To investigate the condition of Chicken parvovirus(ChPV)in chicken small intestine with bleeding lesions in Hebei province,the non structural(NS)protein genes of ChPV in 151 objective samples were amplified by PCR,and genetic evolution of the ChPV NS gene from some positive cases was analyzed.The results of PCR showed that the ChPV positive rate was 48.3%.The positive rates were 64.3%and 44.7%in broilers and layer hens,respectively.Young chicken flocks had more higher positive rates.The more higher positive rates of ChPV were seen in April to May and July to August.The result of sequence alignment revealed that the highest homology of NS gene between 3 isolates and reference strain ChPV/Poland/G090/2011 was from 98.8%to 99.1%,which belonged to the Turkey parvovirus(TuPV)subgroup.And that were both 99.7%between the ChPV/CK/CH/HBZJK/17 isolate and reference strain GX-CH-PV-17 and GX-CH-PV-14,which belonged to the ChPV subgroup.The results illustrated that the detection rate of ChPV was higher in chicken small intestine with bleeding lesions in Hebei province,and the ChPV in the sequenced samples belonged to the ChPV subgroup and the TuPV subgroup,respectively.
