9株四川省成都地区猪繁殖与呼吸综合征病毒ORF5基因的克隆与分析 被引量:3

Cloning and analysis of ORF5 gene of nine strains of Porcine reproductive and respiratory syndrome virus in Chengdu area of Sichuan Province

摘要 为了了解四川省成都地区猪繁殖与呼吸综合征病毒(Porcine reproductive and respiratory syndrome virus, PRRSV)流行毒株的分子生物学特性及遗传变异情况,试验采用生物信息学软件,对从四川省成都地区部分中小型养殖场猪群中收集的9份PRRSV阳性样品进行ORF5基因序列比对及遗传变异分析。结果表明:PCR扩增得到大小约为937 bp的条带,阳性克隆菌的测序结果与GenBank中PRRSV的同源性均大于98%;9株毒株SC20-13、SC20-19、SC20-27、SC20-31、SC20-35、SC20-76、SC20-81、SC20-95、SC21-11的ORF5基因核苷酸序列的同源性为84.4%~100%,其推导氨基酸序列的同源性为83.9%~100%。9株毒株均为PRRSV-2型,其中8株与谱系8的JXA1、CH-1a和CH-1R亲缘关系最近,而1株(SC21-11株)与谱系1的代表株NADC30的亲缘关系最近,SC21-11株GP5蛋白的第33位氨基酸缺失(S/N/G^(33))。说明PRRSV毒株变异频繁,GP5蛋白S/N/G^(33)缺失模式可以演化成为一类新型ORF基因缺失NADC30-like毒株。 In order to understand the molecular and genetic variation of Porcine reproductive and respiratory syndrome virus(PRRSV) in Chengdu area of Sichuan Province, the experiment used bioinformatics software to perform ORF5 gene sequence alignment and genetic variation analysis on nine PRRSV positive samples collected from pig herds of some small and medium-sized farms in Chengdu area of Sichuan Province. The results showed that a band with a size of about 937 bp was obtained by PCR amplification, and the sequencing results of the positive clones were more than 98% homologous to the PRRSV in GenBank. The homology of the ORF5 gene nucleotide sequences of nine sample strains SC20-13, SC20-19, SC20-27, SC20-31, SC20-35, SC20-76, SC20-81, SC20-95, SC21-11 was 84.4%-100%, and the homology of the deduced amino acid sequences was 83.9%-100%. The nine strains were all PRRSV-2 strains, of which eight strains were closely related to JXA1, CH-1 a and CH-1 R, the representative strain of lineage 8, and one strain(SC21-11) was closely related to NADC30, the representative strain of lineage 1. The 33 th amino acid of GP5 protein of the SC21-11 strain had been missing(S/N/G;). The results suggested that PRRSV strains frequently mutated, and the GP5 protein S/N/G;deletion pattern could evolve into a new type of NADC30-like strain with ORF gene deletion.
