摘要
文章通过黑附球菌YL01深层发酵产生胞外黄色素并对黄色素的合成途径进行研究,分析了碳源种类、甘油质量浓度对生物量和产黄色素能力的影响,通过添加次级代谢途径抑制剂来确定黄色素的生物合成途径,并通过添加前体物质对黄色素可能合成途径进行验证。结果表明:以甘油为碳源时可显著提高黑附球菌YL01黄色素的产量,当甘油添加量为20 g/L时,黄色素的产量最高,为15.77 U/mL;莽草酸途径抑制剂三甲胺能够显著降低黄色素的产量;添加莽草酸途径前体物质酪氨酸和苯丙氨酸能促进黄色素合成,酪氨酸浓度为6.0 mmol/L时单位菌体黄色素产量增加88.90%,苯丙氨酸浓度为3.0 mmol/L时单位菌体黄色素产量增加107.83%;黑附球菌YL01胞外黄色素的合成经由莽草酸代谢途径,甘油可促进前体物质合成,从而提高了黄色素产量。
Epicoccum nigrum YL01 was used to produce extracellular yellow pigment(EYP)by submerged fermentation and the possible synthesis pathway of EYP was analyzed.The influences of various carbon sources and glycerol concentration on EYP production and biomass were investigated.Inhibitors of secondary metabolic pathways were added to the broth to determine the possible biosynthetic pathway of EYP and the precursors were introduced to confirm the biosynthetic pathway of EYP.The results showed that the EYP accumulation increased by using glycerol as the carbon source and the maximum EYP content reached 15.77 U/mL when using 20 g/L glycerol.The EYP yield decreased obviously when trimethylamine,an inhibitor of shikimic acid pathway,was added to the broth.The EYP yield increased by 88.90%by the addition of 6.0 mmol/L tyrosine and the EYP yield increased by 107.83%by the addition of 3.0 mmol/L phenylalanine.The synthesis of EYP of Epicoccum nigrum YL01 was through the shikimic acid pathway probably,and glycerol could promote the synthesis of precursor substances and increase the production of yellow pigment.
