摘要 研究了玉米浆硫酸水解制备氨基酸水解液的生产工艺条件。以水解液中氨基酸态氮质量浓度为考核指标,通过单因素试验考察酸料体积比、水解温度和水解时间对玉米浆硫酸水解工艺条件的影响,采用正交试验确定制备玉米浆硫酸水解液的最佳工艺条件。优化后的最佳水解工艺:酸料体积比0.07∶1,水解温度115℃,水解时间12 h。影响玉米浆硫酸水解效果的因素排序是:酸料体积比>水解温度>水解时间。按最优工艺条件制备玉米浆水解液,其氨基酸态氮质量浓度为18.9 g/L,水解率达73.2%。与使用玉米浆为发酵配料相比,使用玉米浆硫酸水解液为温敏型谷氨酸发酵配料可以提高发酵水平的稳定性。 Conditions of a production process was investigated for the preparation of amino acid hydrolysates from corn steep liquor(CSL)by sulphuric acid hydrolysis.The effects of the ratio of acid volume to raw material,hydrolysis temperature and hydrolysis time on the concentration of amino acid nitrogen in the hydrolysate were investigated by single factor and orthogonal experiments.The results demonstrated that the optimal hydrolysis conditions were as follows:the ratio of acid volume to raw material was 0.07∶1,hydrolysis temperature was 115℃and hydrolysis time was 12 h.Factors affecting the sulfuric acid hydrolysis of CSL was in this order:the ratio of acid volume to rawmaterial>hydrolysis temperature>hydrolysis time.Under the optimal hydrolysisconditions,the hydrolyzate contain 18.9 g/L amino acid nitrogen and the protein hydrolysis rate was 73.2%.Compared with CSL,the hydrolysis of CSL produced by this process can improve the stability of fermentation used in temperature-sensitive glutamic acid fermentation compared with CSL.
