固相萃取-超高效液相色谱法测定鱼腥草中的汞

Determination of Mercury in Houttuynia Cordata Thumb.by Ultra-performance Liquid Chromatography after Solid Phase Extraction

摘要 目的:建立了固相萃取净化,超高效液相色谱测定鱼腥草中汞的方法。方法:样品经水浴消解提取后,消解液中汞与二乙基二硫代氨基甲酸钠(DDTC)反应形成络合物,采用多壁碳纳米管固相萃取富集,以甲醇和水为流动相,通过BEH C18柱分离,用二极管阵列检测器检测。结果:在最佳实验条件下,汞的DDTC络合物在3.0~300 μg/L线性范围内,线性关系良好,相关系数(r)大于0.999;方法的检出限为0.15 μg/L,定量限为0.5 μg/L;加标回收率在73.6%~84.8%之间,相对标准偏差(RSD)为5.7%。结论:建立了成本较低、灵敏度高、线性范围宽的超高效液相色谱用于测定汞的方法,为研究金属污染提供新的分析手段。 Objective:A method using solid phase extraction for purification was developed for the determination of mercury in Houttuynia cordata Thumb.by ultra-performance liquid chromatography.Methods:The sample was digested and extracted with a water bath,and then the extractswere reacted with sodium diethyldithiocarbamate (DDTC) to form a complex,which was enriched by solid-phase extraction with multi-walled carbon nanotubes.The complex was performed on a BEH C18 column with methanol and water as mobile phases and detected by photodiode array detected.Results:Under the optimum operation conditions,the DDTC complex of mercury showed good linearity within the linear range of 3.0~300 μg/L,with the correlation coefficients (r) more than 0.999.The limits of detection and quantification were 0.15 μg/L and 0.5 μg/L.The average recoveries deviations ranged from 73.6% to 84.8% with the relative standard deviation (RSD) of 5.7%.Conclusion:A low-cost,high-sensitivity,wide-linear ultra-performance liquid chromatography method for the determination of mercury has been established,which provides a new analytical method for the study of metal pollution.
