摘要
为探讨维生素C(VC)对白斑综合征病毒(White spot syndrome virus,WSSV)侵染后红螯光壳螯虾Cherax quadricarinatus血淋巴免疫机能的影响,在基础饲料中添加不同水平的VC(0(对照)、100、200、400、800、1600、3200 mg/kg),投喂初始体质量为(20.31±2.09)g的红螯光壳螯虾8周,在养殖的第2、4、6、8周测定各组的相关免疫指标,养殖试验结束后在0、400、800 mg/kg VC添加组进行WSSV攻感染试验,测定WSSV侵染后血细胞总数、吞噬活性,以及血淋巴中溶血活性、凝集活性、抑菌率的变化,并分析凝集素基因(C-lectin)表达量的变化。结果表明:WSSV侵染后,红螯光壳螯虾血细胞总数及小颗粒细胞、透明细胞2种类型的血细胞数量均显著减少(P<0.05),而大颗粒血细胞数量则显著增加(P<0.05),至96 h时,800 mg/kg VC添加组血细胞总数显著高于对照组(P<0.05);血淋巴溶血活性、凝集活性和抑菌率随着WSSV侵染时间的延长,各组均呈先升高后降低的趋势,至96 h时,800 mg/kg VC添加组血淋巴凝集活性和抑菌率显著高于对照组(P<0.05);红螯光壳螯虾C-lectin基因在血细胞中表达量最高,显著高于其他组织(P<0.05),至96 h时,试验组血细胞中的C-lectin基因表达量均保持在较低水平,且800 mg/kg VC添加组基因表达量显著高于对照组和400 mg/kg VC添加组(P<0.05)。研究表明,VC能在一定程度上增强红螯光壳螯虾的免疫机能,当添加量为800 mg/kg时,能显著增强血细胞及血淋巴的免疫活性,对红螯光壳螯虾防御WSSV侵染起到一定的免疫保护作用。
To investigate effect of vitamin C(VC)on haemolymph immune function of marron Cherax quadricarinatus infected with white spot syndrome virus(WSSV)by injection of WSSV,marron with initial body mass of(20.31±2.09)g and initial body length of(19.88±3.17)cm were reared in 18 tanks of each 1.0 m×0.5 m×0.5 m at bottom with 4-6 tiles and 10 pieces of PVC tube at stocking rate of 8 individuals and fed diets containing VC at a dose of 0(control),100,200,400,800,1600 and 3200 mg/kg at water temperature of 26℃for 8 weeks.The total hemocyte count,phagocytic activity,and hemolytic activity,hemagglutination activity and bacteriostatic rate were detected in the haemolymph in the marron administrated with WSSV infection in 2,4,6,and 8 weeks and C-lectin gene expression were analyzed in hepatopancreas,gill,muscle,stomach,intestine,brain,and antennal gland using molecular biology technology in the marron administrated with WSSV infection at a dose of 0,400,and 800 mg/kg in 2,4,6,and 8 weeks.The results showed that the number of hemocytes was increased by 42.26%in the marron in the 800 mg/kg VC supplement group compared with the control group(P<0.05),with the maximal agglutination activity and antibacterial rate in hemolymph of the marron fed the diet containing 800 mg/kg VC,significantly higher than those in the control group(P<0.05).Although 96 hours of WSSV infection significantly led to decrease in the total number of hemocytes,phagocytic activity,agglutination activity and antibacterial rate were significantly higher in 800 mg/kg VC addition group at 96 h than those in control group(P<0.05).There was significantly higher expression of C-lectin gene in hemocytes,the maximal expression level of C-lectin gene was observed in hemocytes,than that in other tissues(P<0.05),with significant decrease in C-lectin gene expression in the marron exposed to WSSV infection(P<0.05),and significantly higher gene expression in 800 mg/kg VC addition group than that in control group and in 400 mg/kg VC addition group(P<0.05).The findings indicated that VC enhanced the immune function of marron fed the diet containing 800 mg/kg of witamin C,showing an immune protective effect on marron against WSSV infection.
