肾虚育精方对肾精亏虚少弱精子症小鼠模型精子DNA损害及生精细胞凋亡的干预研究 被引量:2

摘要 目的探讨肾虚育精方对肾精亏虚少弱精子症小鼠模型精子DNA损害及生精细胞凋亡的干预效果。方法通过房劳联合惊恐的伤肾法复制肾精亏虚少弱精子症小鼠模型。动物实验分为正常组、模型组、五子衍宗丸组、肾虚育精方组四组,每组10只。按实验要求饲养父代小鼠以及子代小鼠后,予以取材。采用流式细胞术、Westernblot依次检测父代小鼠的睾丸组织病理损伤、生精细胞凋亡、睾丸组织内Bcl-2/Bax的表达;血球计数法测定子代小鼠附睾内的精子数量。结果与正常组相比,模型组父代小鼠生精细胞凋亡率、睾丸组织Bax的表达量明显升高,Bcl-2的表达量明显下降,子代小鼠附睾内精子浓度明显减少,差异具有统计学意义(P<0.05)。与模型组相比,肾虚育精方组和五子衍宗丸组的生精细胞数量有所增加,生精细胞凋亡率、父代睾丸组织Bax的表达量明显下降,Bcl-2的表达量明显升高,子代小鼠附睾内精子浓度明显增加,差异具有统计学意义(P<0.05)。结论肾虚育精方可通过调节Bcl-2/Bax蛋白的表达,降低肾精亏虚少弱精子症小鼠生精细胞的凋亡率。 Objective To investigate the effect of intervention of Shenxu Yujing Recipe on sperm DNA damage and spermatogenic cell apoptosis in asthenospermia model of kidney deficiency mice. Methods A mouse model of kidney deficiency and asthenospermia was constructed by the method of injury of the kidneys combined with sexual strain. The animal experiment was divided into four groups: normal group, model group, Wuzi Yanzong pill group, and Shenxu Yujing Recipe group, with 10 mice in each group. Materials was collected after the parent mice and their offspring were raised according to the experimental requirements. Flow cytometry, Western blot were used to detect spermatogenic cell apoptosis, the expression of Bcl-2/Bax in the testis tissue. The blood count method was used to determine the sperm count in the epididymis of the offspring mice. Results Compared with the normal group, the apoptosis rate of spermatogenic cells and the expression level of Bax in testicular tissue of the paternal mice in the model group were significantly increased, the expression level of Bcl-2 was significantly decreased, and the sperm concentration in epididymis of the offspring mice was significantly decreased, with statistical significance(P<0.05). Compared with model group, the number of spermatogenic cells in Shenxu Yujing Recipe group and Wuziyanzong Pill group was increased, the apoptosis rate of spermatogenic cells, the expression level of Bax in paternal testicular tissue was significantly decreased, the expression level of Bcl-2 was significantly increased, and the sperm concentration in epididymides of offspring mice was significantly increased, with statistical significance(P<0.05). Conclusions Shenxu Yujing Recipe can reduce the apoptosis rate of spermatogenic cells by regulating the expression of Bcl-2/Bax protein.
