利拉鲁肽对KKAy小鼠脂代谢及PI3K蛋白表达的影响 被引量:1

摘要 目的:探讨利拉鲁肽对KKAy小鼠脂代谢及PI3K蛋白表达的影响。方法:选取12只KKAy小鼠随机分为利拉鲁肽组、模型组,每组各6只;6只C57BL/6J小鼠作为对照组。利拉鲁肽组予小鼠利拉鲁肽注射液(250μg·kg^(-1),ih),对照组和模型组小鼠皮下注射等量生理盐水;所有小鼠持续给药6周。末次给药后,代谢笼测定小鼠进食及饮水量;眼眶取血测总胆固醇(TC)及甘油三酯(TG)水平;小鼠处死后称肝脏重量并计算肝脏指数;苏木素-伊红染色(HE)观察肝脏病理学改变;免疫组化检测肝脏组织PI3K蛋白表达情况。结果:利拉鲁肽可降低KKAy小鼠饮水量、肝脏重量及肝脏指数,提高肝脏PI3K蛋白表达。HE染色结果显示:利拉鲁肽可减轻肝细胞肿胀和炎症程度,减少肝细胞空泡样变,增加细胞核。结论:利拉鲁肽可改善KKAy小鼠脂代谢及脂肪沉积状况,并促进其肝脏PI3K蛋白表达。 Objective:To investigate the effect of liraglutide on lipid metabolism and expression of PI3K protein in KKAy mice.Methods:Twelve KKAy mice were randomly divided into liraglutide group and model group,with 6 mice in each group.Six C57BL/6J mice were used as control group.Mice in the liraglutide group were given liraglutide injection(250μg·kg^(-1),ih).And mice in the control group and model group were given equivalent normal saline subcutaneously.All mice were administered continuously for 6 weeks.After the last administration,the consumption of food and water was measured in the metabolic cage.Orbital blood samples were taken for the detection of the levels of total cholesterol(TC)and triglyceride(TG).After the mice were killed,the weight of the liver was weighed and the liver index was calculated.Hematoxylin eosin staining(HE)was used to observe the pathological changes of the liver.Immunohistochemistry was used to detect the expression of PI3K protein in liver tissue.Results:Liraglutide could significantly reduce the water consumption,liver weight,liver index and increase the expression of PI3K protein in the liver of KKAy mice.HE staining results showed that liraglutide could reduce the degree of swelling and inflammation of hepatocytes,reduce vacuolar degeneration of hepatocytes and increase the number of nuclei.Conclusion:Liraglutide could improve lipid metabolism,lipid deposition and promote the expression of PI3K protein in liver of KKAy mice.
