摘要
用单因素法和正交试验对拮抗菌边缘假单胞菌Pseudomonas marginalis CJT23进行发酵条件优化。单因素条件优化结果表明:环境条件装液量为50%,接种量为1%(体积分数),盐度为1%,pH为7,转速为130 r/min,温度为20℃,其抑菌活性最强;营养条件碳源为葡萄糖、氮源为胰蛋白胨、无机盐为氯化钙、生长因子为半胱氨酸时,抑菌圈直径最大。正交试验结果表明:影响因素大小顺序为碳源>无机盐>氮源>生长因子,即酵母>氯化钠>胰蛋白胨>酪氨酸;发酵时间>盐度>pH,即发酵12 h,pH为6,盐度为3%,抑菌圈直径最大,抑菌活性最强。该结果为拮抗菌P.marginalis CJT23的进一步开发利用提供了理论依据,也为微生物制剂生产提供了参考资料。
The fermentation conditions of antagonistic Pseudomonas marginalis CJT23 were optimized by single factor method and orthogonal test.The results of single factor optimization showed that the best antibacterial activity was obtained under the environmental conditions of 50%liquid loading,1%inoculum size,1%salinity,pH of 7,130 r/min rotation speed and 20℃.The diameter of bacteriostatic circle was the largest when the carbon source was glucose,the nitrogen source was tryptone,the inorganic salt was calcium chloride and the growth factor was cysteine.The results of orthogonal experiment showed that the influencing factors were carbon source>inorganic salt>nitrogen source>growth factor,namely yeast>sodium chloride>tryptone>tyrosine.Fermentation time>salinity>pH,that is,fermentation time is 12 h,pH is 6,salinity is 3%,and the diameter of bacteriostatic circle is the largest,and the bacteriostatic activity is the strongest.The results provide a theoretical basis for further development and utilization of antagonistic bacteria P.marginalis CJT23,and also provide references for the production of microbial agents.
