摘要
目的:探索虫草参多酚提取物对定位和非定位羟基自由基介导的2-脱氧核糖裂解的抑制作用,以期为虫草参新功能的开发和利用提供理论参考。方法:采用Folin-酚法测定虫草参游离酚和结合酚提取物的酚含量,采用体外Fenton反应实验体系,以对2-脱氧核糖裂解的抑制率为考察指标,测定3个采收期2个采收地虫草参多酚提取物对定位和非定位羟基自由基介导的2-脱氧核糖裂解的抑制作用。结果:虫草参游离酚和结合酚提取物的酚含量范围分别为452.10~527.88 μg GAE/mg EDW和140.00~148.46 μg GAE/mg EDW;在非定位羟基自由基介导的体系中,虫草参游离酚和结合酚提取物的IC;值范围分别为2.9627~3.6375 mg/mL和6.1663~10.7542 mg/mL;在定位羟基自由基介导的体系中,虫草参游离酚和结合酚提取物的IC;值范围分别为0.7669~1.2958 mg/mL和1.1067~1.6557 mg/mL。结论:虫草参游离酚和结合酚提取物对定位羟基自由基介导的2-脱氧核糖裂解的抑制作用均强于非定位羟基自由基,虫草参游离酚提取物对羟基自由基介导的2-脱氧核糖裂解的抑制作用强于虫草参结合酚提取物,虫草参多酚提取物对2-脱氧核糖裂解的抑制作用与酚含量之间无显著相关性。
Objective:The inhibitory effect of polyphenol extracts from Lycopus lucidus Turcz.on hydroxyl radical-mediated 2-deoxyribose degradation was studied to provide references for the development and utilization of Lycopus Lucidus Turcz.new functions.Methods:The free phenolic contents and bound phenolic extracts from Lycopus lucidus Turcz.were determined by Folin-phenol method.The inhibitory effect of polyphenol extracts from Lycopus lucidus Turcz.on site and non-site hydroxyl radical-mediated 2-deoxyribose degradation were determined using the Fenton reaction model in vitro and the inhibition rate was used as an indicator.Results:The free phenolic content and bound phenolic extracts from Lycopus lucidus Turcz.were in the range of 452.10~527.88 and 140.00~148.46 μg GAE/mg EDW,respectively.In non-site hydroxyl radical-mediated 2-deoxyribose degradation assay,the IC;values of free and bound phenolic extracts were in the range of 2.9627~3.6375 and 6.1663~10.7542 mg/mL,respectively;In site hydroxyl radical-mediated 2-deoxyribose degradation assay,the IC;values of free and bound phenolic extracts were in the range of 0.7669~1.2958 and 1.1067~1.6557 mg/mL,respectively.Conclusion:The inhibitory effect of polyphenol extracts from Lycopus lucidus Turcz.on site hydroxyl radical-mediated 2-deoxyribose degradation was stronger than that of non-site hydroxyl radical-mediated 2-deoxyribose degradation.The inhibitory effect of free phenolic extracts from Lycopus lucidus Turcz.on hydroxyl radical-mediated 2-deoxyribose degradation was stronger than that of bound phenolics.In addition,there was no significant correlation between phenolic content and hydroxyl radical-mediated 2-deoxyribose degradation.
