同位素内标-超高效液相色谱-串联质谱法同时测定板栗中16种真菌毒素 被引量:8

Simultaneous Determination of 16 Kinds of Mycotoxins in Chestnut Using Isotope Internal Standard-High Performance Liquid Chromatography–Tandem Mass Spectrometry

摘要 目的:建立超高效液相色谱-串联质谱法测定板栗中16种真菌毒素。方法:实验对基质效应进行了分析,比较了不同流动相对分离的影响以及不同色谱柱的分离效果,并对超高效液相色谱-串联质谱仪条件进行了优化。结果:板栗基质会产生基质效应,13种真菌毒素产生基质抑制效应,3种真菌毒素产生基质增强效应,其中6种真菌毒素的基质效应大于20%不能忽略。在最优的流动相条件、最佳的色谱柱和优化的洗脱梯度条件下,16种真菌毒素实现良好分离。该方法回收率为80.0%~118.6%,精密度为2.5%~9.1%,检出限(LOD)为0.10~15 μg/kg。结论:该方法操作简便,回收率、灵敏度、检出限等均满足方法学要求,适用于板栗中16种真菌毒素的测定。 Objective:To establish a method for the determination of 16 kinds of mycotoxins in chestnut samples by ultra high performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS).Methods:Matrix effects of chestnut to 16 mycotoxins were studied,confirmed the better mobile phases system for several different systems,decided which column will be the best.Finally,the instrument conditions of the ultra high performance liquid chromatography-triple quadrupole mass spectrometer were also optimized.Results:The matrix effect was observed in chestnut matrix.13 mycotoxins produced matrix inhibitory effects,and 3 mycotoxins produced matrix enhancing effects,among which the matrix effect of 6 mycotoxins was greater than 20% and could not be ignored.All the 16 mycotoxins were well separated under the optimal conditions of mobile phase,chromatographic column and elution gradient.The precision range was 2.5%~9.1%,the recoveries between 80.0%~118.6% and the limit of detection (LOD) was 0.10~15 μg/kg.Conclusion:This method is simple and rapid,the sensitivity,precision and recovery are all satisfied with the methodological requirements,which can be suitable for determination of 16 kinds of mycotoxinsin in chestnut samples.
