摘要
目的:通过研究钩薄过敏颗粒对咳嗽变异性哮喘(cough variant asthma,CVA)大鼠肺组织基质金属蛋白酶-9(MMP-9)、基质金属蛋白酶抑制剂-1(TIMP-1)mRNA及蛋白表达的影响,探讨其治疗CVA的可能机制。方法:将30只SPF级雄性SD大鼠随机分为正常组、模型组、钩薄过敏颗粒低、高剂量组、地塞米松组;每组6只。除正常组外,其余各组给予10%OVA和Al(OH)3混合物皮下注射建立致敏CVA大鼠模型,第15天开始每日给药,第28天将大鼠处死,RT-PCR方法检测大鼠肺组织MMP-9、TIMP-1 mRNA表达,Western-blot技术检测大鼠肺组织MMP-9、TIMP-1蛋白表达。结果:与正常组相比,模型组肺组织中MMP-9、TIMP-1 mRNA和蛋白水平明显增加(P<0.01);与模型组相比,钩薄过敏颗粒组肺组织中mRNA及MMP-9、TIMP-1蛋白水平明显下降(P<0.01)。结论:钩薄过敏颗粒治疗CVA有效,其作用机制可能与下调肺组织中MMP-9、TIMP-1基因及蛋白表达有关。
Objective:To investigate the mRNA and protein expression of TIMP-1 and MMP-1 in cough variant asthma(CVA)rats treated with Goubo anti-allergy particles,and to determine the mechanism on the therapy.Methods:Thirty male SD rats of SPF grade were randomly divided into normal group,model group,low and high dose groups of Goubo allergic particles and dexamethasone group(6 rats in each group).Except those in the normal group,rats were sensitized with ovalbumin(OVA)and Al(OH)3 to form CVA.On the 15th day,the rats were fed with indicated medicine daily,respectively.After 14 days,the rats were killed to collect lung tissue specimens for tests.The mRNA expressions of MMP-9 and TIMP-1 in lung tissue were detected by RT-PCR,and the protein expressions of MMP-9 and TIMP-1 were detected by Western blot.Results:Compared with the normal group,the protein and mRNA levels of MMP-9 and TIMP-1 in the lung tissue were significantly increased in the model group(P<0.01).Compared with the model group,the protein and mRNA levels of MMP-9 and TIMP-1 in the lung tissue of high dose group were decreased(P<0.01).Conclusion:Goubo anti-allergy particles can treat CVA rats.The mechanism may be related to the down regulation of MMP-9 and TIMP-1 in lung tissue.
