三七不同部位提取物体外抗氧化功效研究 被引量:5

摘要 目的:研究三七不同部位提取物的抗氧化功效,为三七化妆品原料的开发提供实验依据。方法:采用超氧阴离子自由基清除活性实验及羟自由基清除活性实验检测三七根、茎、叶、花提取物对超氧阴离子自由基及羟自由基的清除活性,依据自由基清除率计算半抑制浓度(inhibitory concentration 50,IC50)。采用H;O;诱导人皮肤成纤维细胞(human foreskin fibroblasts, HFF-1)氧化损伤模型,以50 mg·L;的三七根、茎、叶、花提取物分别作用HFF-1细胞,培养24 h后观察细胞形态;MTT法测定细胞活力;微板法检测细胞中超氧化物歧化酶(superoxide dismutase, SOD)的活力及还原型谷胱甘肽(reduced glutathione, GSH)和丙二醛(malondialdehyde, MDA)的含量;酶联免疫吸附测定实验(enzyme linked immunosorbent assay, ELISA)检测细胞中Ⅰ型胶原蛋白(type I collagen, ColⅠ)和ColⅢ的含量。结果:三七根、茎、叶、花提取物对超氧阴离子自由基及羟自由基均具有清除作用;与模型组比较,三七根、茎、叶、花提取物均能不同程度的提升细胞活力及SOD活力(P<0.01),提高GSH及ColⅠ、ColⅢ的水平(P<0.05),降低细胞内MDA的含量(P<0.05),改善氧化损伤模型细胞的形态学变化,且三七根提取物组作用效果最为显著。结论:三七根、茎、叶、花提取物均具有明显的抗氧化作用,其中三七根提取物的抗氧化功效最好。 Objective: To study the antioxidant effect of extracts from different parts of Sanqi(Radix Notoginseng),so as to provide experimental basis for the development of cosmetic raw materials of Sanqi(Radix Notoginseng).Methods: The scavenging activities of Sanqi(Radix Notoginseng) root, stem, leaf and flower extracts on superoxide anion free radical and hydroxyl free radical were detected by superoxide anion free radical scavenging activity test and hydroxyl free radical scavenging activity test.The half inhibitory concentration 50(IC50) was calculated according to the free radical scavenging rate.The oxidative damage model of human foreskin fibroblasts(HFF-1) was induced by H;O;.The HFF-1 cells were treated with 50 mg/L Sanqi(Radix Notoginseng) root, stem, leaf and flower extracts respectively.The cell morphology was observed after 24 hours of culture.Cell viability was measured by MTT assay.The activity of superoxide dismutase(SOD) and the contents of reduced glutathione(GSH) and malondialdehyde(MDA) were detected by microplate method.Enzyme linked immunosorbent assay(ELISA) was used to detect the contents of type Ⅰ collagen(Col Ⅰ) and Col Ⅲ.Results: The extracts from roots, stems, leaves and flowers of Sanqi(Radix Notoginseng) had scavenging effects on superoxide anion radical and hydroxyl radical;compared with the model group, the extracts of Sanqi(Radix Notoginseng) root, stem, leaf and flower could increase the cell activity and SOD activity in varying degrees(P<0.01),increase the levels of GSH,Col Ⅰ and Col Ⅲ(P<0.05),reduce the content of intracellular MDA(P<0.05),and improve the morphological changes of oxidative damage model cells, and the effect of Sanqi(Radix Notoginseng) root extract group was the most significant.Conclusion: Sanqi(Radix Notoginseng) root, stem, leaf and flower extracts have obvious antioxidant effect, and Sanqi(Radix Notoginseng) root extract has the best antioxidant effect.
