摘要
健康长寿是人类一直追求的目标,作为复杂的生物学现象,其与复杂的遗传和环境因素的联合作用密切相关。我国老龄化形势严峻,长寿而不健康的问题依然突出。近年来,我国相关研究机构针对健康长寿的关键影响因素进行了研究,取得了一定进展。本文阐述中国健康长寿纵向调查队列(CLHLS)、广西自然长寿队列、海南自然长寿人群等主要的健康长寿人群资源,综述我国科学挖掘的影响人群健康长寿的遗传基因、长链非编码RNA及甲基化表观遗传修饰和线粒体含量等领域的最新进展,并针对性别、社会、心理健康、营养、肠道菌群等研究热点与健康长寿的关系进行分析,对未来的健康长寿研究提出建议。从临床医学、公共卫生、基础医学、社会学等多维度系统进行整合,对人类表型组做全面的介绍,精确测量、精密解析"基因—环境—表型"之间的多层次关联及整体性关系,以科技为健康老龄化的战略实施赋能。
Healthy longevity is the goal that human beings have been pursuing.As a complex biological phenomenon,it is closely related to the combined effect of complex genetics and environmental factors.The aging problem in China is serious,and the challenge of unhealthy longevity is still prominent.Both of genetic background or environmental,lifestyle and social factors,we are significantly different from Western countries.Human are more complex than animal models with more influencing factors.In recent years,based on the resources of healthy longevity populations,relevant research institutions in China have conducted research on the key influencing factors of healthy longevity and have made certain progress.Here,we summarized the main healthy longevity population resources such as the Chinese Longitudinal Healthy Longevity Survey(CLHLS),the Guangxi Longevity Cohort,and the Hainan Longevity Population,especially reviewed the progress of the longevity genes,long-chain non-coding RNAs,and methylation epigenetic modification and mitochondrial content.Additionally,the relationship between hotspots such as gender,society,mental health,nutrition,and intestinal flora,and healthy longevity were summarized.Future research also requires the integration of multi-dimensional systems such as clinical medicine,public health,basic medicine,sociology,etc.,to have a comprehensive and phenomic understanding of human longevity,and accurately measure and precisely analyze the network of"gene-environment-phenotype"to provide technological empowerment for the implementation of the healthy aging strategy.
