骨转移肺鳞癌的血浆长链非编码RNA差异表达分析与MIR4697HG预测价值初步验证 被引量:1

摘要 目的鉴定并验证血浆MIR4697HG对肺鳞癌(LUSC)患者骨转移以及预后的预测能力。方法回顾性收集LUSC患者58例,其中无骨转移41例(均无其他部位转移),骨转移17例(单处骨转移5例,多处骨转移12例)。最常见转移部位为肋骨(8例,47.1%),合并其他部位转移13例(76.4%)。采集患者的血浆标本,选择3对骨转移与非骨转移血浆进行lncRNA测序,筛选出MIR4697HG,进一步在58例血浆样本中进行骨转移预测效能验证。结果lncRNA测序并分析后筛选出456个差异lncRNA,其中MIR4697HG的P值最低。qPCR验证结果提示,MIR4697HG具有良好的预测价值。泛癌分析发现,MIR4697HG在多种肿瘤中呈低表达,可能行使广泛的抑癌基因角色。进一步分析发现,MIR4697HG的表达随着LUSC分期的升高逐渐下降,MIR4697HG低表达的肺癌患者,无病生存率和总生存率均较低。结论MIR4697HG是一个普遍抑癌分子,血浆MIR4697HG可作为LUSC骨转移的相关无创标记物,低表达与LUSC患者不良肿瘤学预后有关。 Objective To identify and verify the predictive ability of plasma MIR4697HG for bone metastasis and prognosis in patients with lung squamous cell carcinoma.Methods Plasma samples were collected from 58 patients with lung squamous cell cancer,including 41 cases without bone metastasis(no other metastasis),17 cases of bone metastasis(5 cases of single bone metastasis,12 cases of multiple bone metastases).The most common metastasis site was ribs(8 cases,47.1%),13 cases(76.4%)combined with other metastases.Three pairs of plasma samples of lung squamous cell carcinoma with and without bone metastasiswere selected for lncRNA sequencing,to screen for MIR4697HG.Then validation of the predictive power for bone metastasis was examined in all 58 plasma samples.Results A total of 456 differentially expressed lncRNAs were screened after sequencing and analysis,among which MIR4697HG had the lowest p value.The results of qPCR showed that MIR4697HG had good predictive value.Pan cancer analysis showed that MIR4697HG was low expressed in a variety of cancers,which might play a wide range of tumor suppressor gene roles.Further analysis showed that the expression of MIR4697HG decreased with the increase of lung squamous cell carcinoma stage.Lower expression of MIR4697HG also was related to lower disease-free survival rate and lower overall survival rate of lung squamous cell carcinoma.Conclusion The plasma MIR4697HG can be used as a noninvasive marker for bone metastasis of lung squamous cell carcinoma.MIR4697HG is a universal tumor suppressor,and low expression of MIR4697HG is associated with poor oncological prognosis in patients with lung squamous cell carcinoma.
