基于Nfr2/HO-1信号通路探讨清肺止咳方对急性支气管炎模型小鼠的影响 被引量:4

摘要 目的:基于抗氧化应激角度探讨Nfr2/HO-1信号通路对急性支气管炎的作用机制,验证清肺止咳方对急性支气管炎的疗效.方法:将80只SPF级小鼠随机分为模型组(64只)、正常组(16只),采用烟熏法制备急性支气管炎模型小鼠,烟熏1周后,随机从两组中各选4只小鼠,取其肺组织进行病理形态学检测,进行模型鉴定.造模成功后,将模型组随机平均分为5组:空白模型组,小儿清热止咳口服液阳性对照组,清肺止咳方低、中、高剂量组;正常组、空白模型组给予生理盐水灌胃,余组予相应药物灌胃治疗,共计1周.治疗结束后,运用免疫组化法测定全部小鼠肺组织IL-1β、IL-6的平均积分光密度,蛋白免疫印迹法(Western blot)测定全部小鼠肺组织中Nfr2、HO-1蛋白表达水平.结果:与正常组相比,空白模型组IL-1β、IL-6积分光密度升高(P<0.05),Nfr2、HO-1蛋白表达水平下降,均具有统计学差异(P<0.05).与空白模型组相比,各药物组IL-1β、IL-6积分光密度均降低(P<0.05);Nfr2、HO-1蛋白表达水平均升高(P<0.05);与阳性对照组相比,清肺止咳方低、中、高剂量组IL-1β、IL-6积分光密度均下降(P<0.05),Nfr2、HO-1蛋白表达水平均升高(P<0.05);清肺止咳方低中高组间比较,IL-1β、IL-6的平均积分光密度及Nfr2、HO-1蛋白表达水平无统计学差异.结论:Nfr2/HO-1信号通路在急性支气管炎的炎症反应过程中发挥着重要作用,清肺止咳方可能可通过调节该信号通路,促进Nfr2、HO-1表达,抑制IL-1β、IL-6的表达,进而治疗急性支气管炎. Objective:To explore the role of Nfr2/HO-1 signaling pathway in acute bronchitis from the point of antioxidant stress,and to verify the efficacy of Qingfei Zhike Decoction in the treatment of acute bronchitis.Methods:Totally 80 SPF mice were randomly divided into normal group(n=16)and model group(n=64)with smoke-induced acute bronchitis.One week after modeling,4 mice were randomly selected from each group,and their lung tissues were taken for pathomorphological examination and model identification.As successful modeling,the model group was randomly and equally divided into 5 groups:blank model group,positive control group of Xiaoer Qingre Zhike oral liquid,low,middle and high dose groups of Qingfei Zhike Decoction.The normal group and the blank model group was given normal saline,and the other groups were treated with corresponding drugs for a total of one week.After treatment,the average integral optical density of IL-1βand IL-6 in lung tissue of all mice was measured by immunohistochemical method,and the expression of Nfr2 and HO-1 protein in lung tissue of all mice was determined by Western blotting.Results:Compared with the normal group,the integrated optical density of IL-1βand IL-6 in the blank model group increased(P<0.05),while the expression of Nfr2 and HO-1 decreased(P<0.05).Compared with the blank model group,the integral optical density of IL-1βand IL-6 decreased(P<0.05)and the expression of Nfr2 and HO-1 increased in all drug groups(P<0.05).Compared with the positive control group,the integral optical density of IL-1βand IL-6 in the low,middle and high dose Qingfei zhike Decoction.groups decreased,while the expression level of Nfr2 and HO-1 protein increased(P<0.05).There was no significant difference in the average integral optical density of IL-1βand IL-6 and the expression level of Nfr2 and HO-1 protein between the low,middle and high groups of Qingfei Zhike Decoction(P>0.05).Conclusion:Nfr2/HO-1 signal pathway plays an important role in the inflammatory process of acute bronchitis.Qingfei Zhike decoction may increase the expression of Nfr2 and HO-1,while inhibit the expression of IL-1βand IL-6 by Nfr2/HO-1 signaling pathway to treat acute bronchitis.
