芬太尼通过PI3K/Akt信号通路调控宫颈癌细胞增殖迁移 被引量:2

Regulation of proliferation and migration of cervical cancer cells by fentanyl through PI3K/Akt signaling pathway

摘要 目的探究芬太尼对宫颈癌细胞增殖迁移的作用及分子机制。方法将宫颈癌细胞系C33A分为对照组和芬太尼组(使用不同浓度芬太尼处理),分别采用CCK-8、Transwell法、流式细胞术检测芬太尼对C33A细胞增殖、迁移及凋亡的影响。Western blot法检测活化Caspase 3、PARP1含量,MMP-2、MMP-9、N-cadherin表达水平及PI3K/Akt磷酸化情况。结果相比于对照组,芬太尼显著抑制C33A细胞增殖、迁移,且呈剂量依赖性(P<0.05)。芬太尼组的细胞凋亡率及活化Caspase 3、PARP1含量显著高于对照组,且呈剂量依赖性(P<0.05);相比于对照组,芬太尼组细胞MMP-2、MMP-9、N-cadherin表达水平及PI3K、Akt磷酸化水平显著降低,且呈剂量依赖性(P<0.05)。结论芬太尼具有抑制宫颈癌细胞增殖迁移,并促进宫颈癌凋亡的能力,其作用机制与抑制PI3K/Akt通路激活有关。 Objective:To explore the effect and molecular mechanism of fentanyl on proliferation and migration of cervical cancer cells.Methods:Cervical cancer cell lines C33A were divided into control group and fentanyl group.C33A cells in the fentanyl group were treated with different concentrations of fentanyl.CCK-8,transwell method,scratch test and flow cytometry were used to detect the effects of fentanyl on the proliferation,migration and apoptosis of C33A cells.Western blot was used to detect the levels of activated Caspase 3 and PARP1,the expression levels of MMP-2,MMP-9,N-cadherin,the phosphorylation of PI3K/Akt.Results:Compared with the control group,fentanyl intervention significantly inhibited the proliferation and migration of C33A cells in a dose-dependent manner(P<0.05).The apoptosis rate and the contents of activated Caspase 3 and PARP1 in fentanyl group were significantly higher than those in blank group,in a dose-dependent manner(P<0.05).Compared with the control group,the expression levels of MMP-2,MMP-9,N-cadherin,the phosphorylation levels of PI3K and Akt in fentanyl group were significantly decreased in a dose-dependent manner(P<0.05).Conclusion:Fentanyl can inhibit the proliferation and migration of cervical cancer cells and promote the apoptosis of cervical cancer cells,and its mechanism was related to the inhibition of PI3K/Akt pathway activation.
